目的 探讨低分子量多肽-2(LMP-2)基因多态与类风湿关节炎(RA)的相关性。方法 用多聚酶链反应-限制性片段长度多态性(PCR-RFLP)分析技术检测86例RA患者和80名正常人LMP-2第三外显子基因多态性,同时扩增DRB1基因第二外显子以筛选DR4阳性标本。结果 LMP-2RH杂合子及LMP-2RR纯合子频率在RA组分别显著高于和低于正常对照组(P均<0.05),LMP-2H可使DR4+正常人患RA的相对危险度(RR)增加2.99倍(P<0.05)。结论 LMP-2RH频率升高及LMP-2RR频率降低可能是RA的特征之一,LMP-2H可能与汉族人群RA有一定的关联。

목적 탐토저분자량다태-2(LMP-2)기인다태여류풍습관절염(RA)적상관성。방법 용다취매련반응-한제성편단장도다태성(PCR-RFLP)분석기술검측86례RA환자화80명정상인LMP-2제삼외현자기인다태성,동시확증DRB1기인제이외현자이사선DR4양성표본。결과 LMP-2RH잡합자급LMP-2RR순합자빈솔재RA조분별현저고우화저우정상대조조(P균<0.05),LMP-2H가사DR4+정상인환RA적상대위험도(RR)증가2.99배(P<0.05)。결론 LMP-2RH빈솔승고급LMP-2RR빈솔강저가능시RA적특정지일,LMP-2H가능여한족인군RA유일정적관련。
Objective To evaluate the relationship between polymorphism inlow molecular weight polypeptides-2 (LMP-2) gene and rheumatoid arthritis (RA) in Han nationality.Method A HhnI restriction site polymorphism in the third exon of LMP-2 gene was detected by PCR-RFLP in 86 RA patients and 80 normal controls.Meanwhile,the second exon of DRB1 gene was amplified for screening positive DR4 individuals in the same ethnicity.Results The LMP-2RH heterozygosity and LMP-2RR homozygosity increased and decreased significantly in RA patients respectively compared to normal controls.The LMP-2H genotype could increase the susceptibility to RA for positive DR4 individuals (RR=2.99,P<0.05).Conclusion The increased LMP-2RH and decreased LMP-2RR may be one of the characteristics for RA and the association of LMP-2H with RA may exist in Han nationality.
