研究重组类人胶原蛋白Ⅱ(RHLCⅡ)在CM52树脂上的吸附平衡,考察pH值、NaCl浓度及温度对吸附平衡的影响.结果表明:当4.0<pH≤5.0时,随着pH值增大,吸附容量减小,解离系数K_d增大;当3.0≤pH<4.0时,随着pH值增大,吸附容量增大,解离系数Kd减小;随着NaCl浓度增大,吸附容量减小,解离系数K_d增大;温度对Langmuir方程参数的影响较小,温度每升高10 ℃,吸附容量增加3%左右,解离系数K_d的变化不显著.RHLCⅡ在CM52树脂上的吸附基本符合Langmuir方程,为离子交换法规模制备RHLCⅡ提供了理论依据.

The ion exchange adsorption equilibrium for Recombinant Human-Like Collagen Ⅱ (RHLCⅡ) onto resin CM52 was investigated under different pH values, NaCl concentrations and temperatures. The results show that the dissociation constant K_d is higher and the binding capacity is lower when the pH value increases within 4.0<pH≤5.0. However, when pH value increases within 3.0≤pH<4.0, K_d is lower and the binding capacity is higher. The binding capacity decreases and K_d increases with the increasing of NaCl concentration. Temprature has less influence on Langmuir model parameters, and higher temperature leads to higher binding capacity, increased by 3% per 10 ℃, while no obvious change is observed on K_d. The adsorption of RHLCⅡ onto CM52 obeys Langmuir model, which provides theoretical gist for the large scale production of RHLCⅡ by ion exchange process.
