[目的]探讨大田软海绵酸(okadaic acid,OA)对HL-7702肝细胞的毒性效应.[方法]通过细胞计数试剂盒(Cell Counting Kit-8,CCK-8)、细胞形态学观察及流式细胞术等方法,从细胞增殖、细胞凋亡等方面探讨OA对HL-7702肝细胞的毒性效应.[结果]与溶剂对照组相比,OA对HL-7702肝细胞增殖有明显的抑制作用,细胞增殖抑制率随着OA浓度的增加而增加,同时随着染毒时间的延长,细胞增殖抑制率也明显增加;与对照组相比,作用HL-7702肝细胞24 h后,5 nmol/L OA可明显诱导细胞发生凋亡.[结论]OA可以通过抑制细胞增殖及诱导细胞凋亡的方式对HL-7702肝细胞产生毒性效应,且呈现剂量-效应关系.

[목적]탐토대전연해면산(okadaic acid,OA)대HL-7702간세포적독성효응.[방법]통과세포계수시제합(Cell Counting Kit-8,CCK-8)、세포형태학관찰급류식세포술등방법,종세포증식、세포조망등방면탐토OA대HL-7702간세포적독성효응.[결과]여용제대조조상비,OA대HL-7702간세포증식유명현적억제작용,세포증식억제솔수착OA농도적증가이증가,동시수착염독시간적연장,세포증식억제솔야명현증가;여대조조상비,작용HL-7702간세포24 h후,5 nmol/L OA가명현유도세포발생조망.[결론]OA가이통과억제세포증식급유도세포조망적방식대HL-7702간세포산생독성효응,차정현제량-효응관계.
[Objective] The research aimed to study the cytotoxicity of okadaic acid on HL-7702. [Method] The toxic effect of OA on HL-7702 liver cells was investigated from the aspects of cell proliferation and apoptosis by using Cell Counting Kit-8, cell morphology observation and flow cytometry. [Result] Compared with the control group, OA inhibited the proliferation of HL-7702 liver cells significantly, and the inhibition ratio of cell proliferation increased along with the increase of OA concentration and exposure time. Compared with the control group, 5 nmol/L OA could induce apoptotic cells significantly. [Conclusion] OA exhibited a dose-dependent cytotoxic effect on HL-7702 liver cells by the inhibition of cell proliferation and induction of apoptosis.
