目的通过检测含药血清培养的人乳腺癌细胞中核受体共激活因子(steroid receptor coactivator-1,SRC-1)和核受体共抑制因子(nuclear receptor corepressor,NCOR)的mRNA表达,从分子水平上探讨补肾中药对核受体共调节因子表达的影响.方法 Wistar大鼠随机分为3个大组,每大组再分雌雄2组,每大组分别用淫羊藿、女贞子、蒸馏水灌胃14 d,制备含药血清,并用含药血清培养雌激素受体缺失的乳腺癌细胞MDA-MB-231,利用实时荧光定量RT-PCR方法检测SRC-1和NCOR的mRNA表达.结果补肾阳中药淫羊藿雄性大鼠含药血清上调了SRC-1的表达,对NCOR没有显著影响;而补肾阴中药女贞子下调了SRC-1共激活因子的表达,上调了NCOR的表达.结论补肾中药淫羊藿、女贞子对核受体共调节因子的表达有调节作用.

Objective To explore the effect of different tonifying kidney on expressions of steroid receptor coactivator-1 (SRC-1) and nuclear receptor repressor (NCOR) mRNA. Methods Wistar rats were divided into 3 groups and each group was divided into male and female subgroups. Wistar rats were intragastric administration with epimedium, fructus ligustri lucidi and distilled water for 14 days to make drug serum. Human breast carcinoma cell (MDA-MB-231) was cultured with drug serum and the expressions of SRC-1 and NCOR mRNA were detected. Results Tonifying kidney yang Chinese medicine epimedium could upregulate the expression of SRC-1 in male rats and had no effect on the expression of NCOR. Tonifying kidney yin Chinese medicine fructus ligustri lucidi could downregulate the expression of SRC-1 and upregulate the expression of NCOR. Conclusions Tonifying kidney Chinese medicine epimedium and fructus ligustri lucidi has regulation effect on the expression of steroid receptor coregulator.
