通过比较鸡IgG与鹅IgG的相似性,探讨羊抗鸡酶标二抗用于检测鹅血清抗体的可行性.用辛酸-硫酸铵盐析法提纯鸡、鸭、鹅、小鼠、免、猪、羊的IgG,比较上述动物IgG粗提物的SDS-聚丙烯酰胺凝胶电泳图谱,并用蛋白免疫转印法定性检测上述动物血清IgG与羊抗鸡酶标二抗的结合情况;然后用直接ELISA方法分析不同动物血清分别与抗鸡酶标抗体和抗鹅酶标抗体结合程度的差异;最后用间接EUSA方法比较两种酶标抗体对同一鹅群大肠杆菌抗体的检测结果.结果表明,鸡、鸭、鹅IgG具有较大的相似性,都能够与抗鸡酶标二抗结合;羊抗鸡酶标二抗可以替代抗鹅酶标二抗来检测鹅的血清抗体.

The mechanism of detecting antibody levels in goose serum samples with goat-anti-chicken IgG was clarified by comparing the similarity of chicken IgG with goose IgG.Chicken IgG,duck IgG,goose IgG,mouse IgG,rabbit IgG,pig IgG and goat IgG were salted out by octanoic acid-saturated ammonium sulfate.Their SDS-PAGE pattern was analyzed and the binding was qualitative checked up through western blotting.The binding difference of chicken IgG,duck IgG,goose IgG,mouse IgG,rabbit IgG,pig IgG and goat IgG to anti-chicken-IgG and anti-goose-IgG,were assayed by direct ELISA.Finally the binding difference of two different secondary antibodies to blood-serum of the same group was assayed by indirect ELISA.The results showed that there was much resemblance between chicken IgG,duck IgG and goose IgG,they could also bind to anti-chicken-IgG;Goat-anti-chicken secondary antibody could replace anti-goose IgG to diagnose detecting antibody levels in goose serum.
