目的 探讨高压氧对缺氧缺血性脑损伤(hypoxia-ischemia brain damage,HIBD)大鼠神经细胞凋亡以及炎性细胞因子水平的影响.方法 成年雄性SD大鼠42只,随机分为对照组(6只)、空气加压组(18只)和高压氧(hyperbaric oxyen,HBO)治疗组(18只).采用Pulsineli法制作脑缺血大鼠模型.HBO治疗组第1、3、5天分别给予HBO治疗,1次/d.应用TUNEL染色检测大鼠海马神经元数量,同时检测大鼠细胞因子TNF、IL-1β和IL-6水平.结果 HBO治疗能有效抑制脑缺血引起的细胞因子TNF、IL-1β、IL-6释放(P<0.05或0.01),并减轻大鼠海马区神经元损伤,组间比较差异有统计学意义(P<0.05).结论 HBO治疗可以有效抑制炎性细胞因子的释放,从而保护脑缺血后神经细胞.

Objective To investigate effect of hyperbaric oxygen (HBO) on inflammatory cytokine and neuronal apoptosis in rats with hypoxia ischemia brain damage (HIBD). Methods Forty-two adult male Sprague-Dawley rats were randomly divided into 7 groups (n=6 respectively) following ischemia-hypotension:the blank control group, the ischemia-hypotension 1, 3 and 5 d groups, and the HBO treatment 1, 3 and 5 d groups. The global I/R model was developed with Pulsineli 4 vessel occlusion method. Rats in the HBO treatment groups were exposed to HBO for 1 h a day for a duration of 1 to 5 days. TUNEL staining was used for neuron counting and ELISA was employed to detect levels of TNF, IL-1β and IL-6 in serum. Results Levels of TNF, IL-1β and IL-6 in the HBO treatment groups were lower than those in the untreated groups. After 5 days of HBO treatment, number of neurons in the hippocampal area was significantly higher than that in the untreated group (P <0.05). Conclusions HBO could decrease levels of TNF, IL-1β and IL-6 induced by cerebral ischemia,which reduces the loss of hippocampal neurons after global ischemia-hypotension.
