目的 探讨肠淋巴液引流对失血性休克大鼠液体复苏后肺组织细胞凋亡的作用.方法 18只SPF级Wistar雄性大鼠随机分成假手术组(仅麻醉与手术)、休克组(复制失血性休克模型)、休克+引流组(复制失血性休克模型,并于休克1h后引流休克肠淋巴液)各6只,采用末端脱氧核糖转移介导的生物素化脱氧尿嘧啶缺刻标记法测定肺组织细胞凋亡,细胞核呈棕色的细胞为凋亡细胞,应用免疫组化链酶亲合素-生物素-过氧化物酶复合物法测定bcl-2和bax蛋白表达.结果 休克组与休克+引流组肺组织细胞凋亡率均显著高于假手术组,但休克+引流组肺组织细胞凋亡率显著低于休克组.假手术组bcl-2、bax蛋白均有一定的表达;休克组肺组织细胞bcl-2表达显著低于假手术组,bax表达明显高于假手术组;休克+引流组肺组织细胞bcl-2表达增强,bax表达降低,且休克+引流组肺组织细胞的bcl-2表达显著高于休克组,bax表达显著低于休克组.结论 肺组织细胞凋亡过度是休克后肺损伤的发生机制之一;引流休克肠淋巴液可减少肺组织细胞的凋亡,其作用机制与调节bcl-2/bax蛋白表达有关.
目的 探討腸淋巴液引流對失血性休剋大鼠液體複囌後肺組織細胞凋亡的作用.方法 18隻SPF級Wistar雄性大鼠隨機分成假手術組(僅痳醉與手術)、休剋組(複製失血性休剋模型)、休剋+引流組(複製失血性休剋模型,併于休剋1h後引流休剋腸淋巴液)各6隻,採用末耑脫氧覈糖轉移介導的生物素化脫氧尿嘧啶缺刻標記法測定肺組織細胞凋亡,細胞覈呈棕色的細胞為凋亡細胞,應用免疫組化鏈酶親閤素-生物素-過氧化物酶複閤物法測定bcl-2和bax蛋白錶達.結果 休剋組與休剋+引流組肺組織細胞凋亡率均顯著高于假手術組,但休剋+引流組肺組織細胞凋亡率顯著低于休剋組.假手術組bcl-2、bax蛋白均有一定的錶達;休剋組肺組織細胞bcl-2錶達顯著低于假手術組,bax錶達明顯高于假手術組;休剋+引流組肺組織細胞bcl-2錶達增彊,bax錶達降低,且休剋+引流組肺組織細胞的bcl-2錶達顯著高于休剋組,bax錶達顯著低于休剋組.結論 肺組織細胞凋亡過度是休剋後肺損傷的髮生機製之一;引流休剋腸淋巴液可減少肺組織細胞的凋亡,其作用機製與調節bcl-2/bax蛋白錶達有關.
목적 탐토장림파액인류대실혈성휴극대서액체복소후폐조직세포조망적작용.방법 18지SPF급Wistar웅성대서수궤분성가수술조(부마취여수술)、휴극조(복제실혈성휴극모형)、휴극+인류조(복제실혈성휴극모형,병우휴극1h후인류휴극장림파액)각6지,채용말단탈양핵당전이개도적생물소화탈양뇨밀정결각표기법측정폐조직세포조망,세포핵정종색적세포위조망세포,응용면역조화련매친합소-생물소-과양화물매복합물법측정bcl-2화bax단백표체.결과 휴극조여휴극+인류조폐조직세포조망솔균현저고우가수술조,단휴극+인류조폐조직세포조망솔현저저우휴극조.가수술조bcl-2、bax단백균유일정적표체;휴극조폐조직세포bcl-2표체현저저우가수술조,bax표체명현고우가수술조;휴극+인류조폐조직세포bcl-2표체증강,bax표체강저,차휴극+인류조폐조직세포적bcl-2표체현저고우휴극조,bax표체현저저우휴극조.결론 폐조직세포조망과도시휴극후폐손상적발생궤제지일;인류휴극장림파액가감소폐조직세포적조망,기작용궤제여조절bcl-2/bax단백표체유관.
Objective To explore the effect of the intestinal lymph drainage on lung tissue cell apoptosis in rats with hemorrhagic shock after resuscitation,rich ALI intestinal lymphatic pathway theory.Methods TUNEL method was used to determine the apoptosis of lung tissue cells,the brown nuclei were apoptotic cells.SABC was used to determine Bcl-2 and Bax protein expression.Results The shock group and shock + drainage group lung tissue cell apoptosis rate were significantly higher than those in the sham operation group,but the shock + drainage group,the apoptosis of lung tissue cells was significantly lower than the shock group.Sham operation group showed Bcl-2,Bax protein of expression; In shock group,lung tissue cell Bcl-2 expression was significantly lower than the sham operation group,the Bax expression was significantly higher than that in the sham operation group; In shock + drainage group,lung tissue cells shored enhanced expression of Bcl-2,Bax expression was reduced,and the shock + drainage group lung tissue cell Bcl-2 expression was significantly higher than that in the shock group,the expression of Bax was significantly lower than the shock group.Conclusion The excessive apoptosis of lung tissue cells was one of the mechanisms of lung injury after shock.Intestinal lymph drainage could reduce lung tissue cell apoptosis,the mechanism invdved the regulation of Bcl-2/Bax protein expression.
