目的 观察兔异体冻干跟腱重建前交叉韧带(ACL)后移植物Ⅰ型及Ⅲ型胶原的表达.方法 应用兔异体冻干跟腱重建ACL,手术后3、12、26、52周取移植物,行组织学观察,并应用免疫荧光共聚焦技术(CLSM)检测移植物Ⅰ型及Ⅲ型胶原表达的平均荧光值.结果 术后52周,异体移植物组织形态与正常ACL基本一致;正常ACL细胞周围主要表达I型胶原(57.10±5.04),几乎不表达Ⅲ型胶原(6.09±1.34);术后3周,移植物Ⅲ型胶原阳性表达开始增加,12~26周达到高峰,移植后52周仍有Ⅲ型胶原阳性表达(13.17±2.26),与ACL比较差异有统计学意义(P<0.05);术后3周,移植物Ⅰ型阳性表达开始减少,12周后逐渐增加,52周Ⅰ型胶原阳性表达(52.69±2.76)仍与正常ACL比较差异有统计学意义(P<0.01).结论 异体跟腱重建ACL后,重建后移植物组织形态与正常ACL基本一致,但移植物胶原基质组成与正常ACL存在明显差异.
目的 觀察兔異體凍榦跟腱重建前交扠韌帶(ACL)後移植物Ⅰ型及Ⅲ型膠原的錶達.方法 應用兔異體凍榦跟腱重建ACL,手術後3、12、26、52週取移植物,行組織學觀察,併應用免疫熒光共聚焦技術(CLSM)檢測移植物Ⅰ型及Ⅲ型膠原錶達的平均熒光值.結果 術後52週,異體移植物組織形態與正常ACL基本一緻;正常ACL細胞週圍主要錶達I型膠原(57.10±5.04),幾乎不錶達Ⅲ型膠原(6.09±1.34);術後3週,移植物Ⅲ型膠原暘性錶達開始增加,12~26週達到高峰,移植後52週仍有Ⅲ型膠原暘性錶達(13.17±2.26),與ACL比較差異有統計學意義(P<0.05);術後3週,移植物Ⅰ型暘性錶達開始減少,12週後逐漸增加,52週Ⅰ型膠原暘性錶達(52.69±2.76)仍與正常ACL比較差異有統計學意義(P<0.01).結論 異體跟腱重建ACL後,重建後移植物組織形態與正常ACL基本一緻,但移植物膠原基質組成與正常ACL存在明顯差異.
목적 관찰토이체동간근건중건전교차인대(ACL)후이식물Ⅰ형급Ⅲ형효원적표체.방법 응용토이체동간근건중건ACL,수술후3、12、26、52주취이식물,행조직학관찰,병응용면역형광공취초기술(CLSM)검측이식물Ⅰ형급Ⅲ형효원표체적평균형광치.결과 술후52주,이체이식물조직형태여정상ACL기본일치;정상ACL세포주위주요표체I형효원(57.10±5.04),궤호불표체Ⅲ형효원(6.09±1.34);술후3주,이식물Ⅲ형효원양성표체개시증가,12~26주체도고봉,이식후52주잉유Ⅲ형효원양성표체(13.17±2.26),여ACL비교차이유통계학의의(P<0.05);술후3주,이식물Ⅰ형양성표체개시감소,12주후축점증가,52주Ⅰ형효원양성표체(52.69±2.76)잉여정상ACL비교차이유통계학의의(P<0.01).결론 이체근건중건ACL후,중건후이식물조직형태여정상ACL기본일치,단이식물효원기질조성여정상ACL존재명현차이.
Objective To observe the morphological changes and the expression of Type Ⅰ and Ⅲcolbert in allograft freeze-dried achilles after anterior crueiate ligament(ACL)reconstruction(ACL)in rabbits.Methods The ACLs of rabbits were removed and reconstructed bv allograft freeze-dried achilles.The grafts were removed respectively at the 3rd.12th.26th and 52nd week after surgery.All specimens were stained by hematoxylin and eosin(HE).The confocal laser scanning microscopy(CIJsM)was used to detect the expression of type Ⅰ and Ⅲ collagen.Results Allograft freeze-dried achilles appeared nearly resemble as normalACL at 52weeks after surgeryunderHE stained.As the grafts were observed under confocal laser scanning microscopy,In normal ACL mainly expressed type Ⅰ collagen(57.10±5.04),the expression of typeⅢcollagen(6.09±1.34)nearly none.While in angraft freeze-dried achilies after 52 weeks,there were more expressions of type Ⅲ collagen(13.17±2.26).Moreover,the expressions of type Ⅲ collagen were significantly greater than that of ACL(P<0.05),the expressions of type Ⅰ collagen (52.69±2.76)were significantly less than that of ACL(P<0.01).Conclusion After ACL reconstruction with aligraft freeze-dried achilles,even the morpholagical of the grafts seem as normal ACL but the graftstill did not appear tO the normal ligament structure.
