背景:人发角蛋白人工腱(human hair keratin artificial tendon,HHKAT)材料是国内研制的新型生物可降解材料.目的:研究HHKAT材料在体内的降解及其对周围组织的影响.设计:完全随机双盲实验对照研究.地点和对象:在解放军昆明总医院动物实验基地,选用日本大耳白兔(普通级)12只,体质量(2.5±0.5)kg,雄性,标准饲料单笼喂养.干预:将12只兔随机分组,在脊旁肌埋藏处理30 min和处理15 min的人发角蛋白人工腱试件,用正常人发作对照.分别在2,6,12,24周取材.主要观察指标:对不同时期的材料进行大体及组织学观察.结果:实验中发现不同程度处理的HHKAT其降解速度不同,其中降解最快的处理30min组在24周时已完全吸收,而处理15 min组在24周时只有部分降解,对照组未见降解.HHKAT及人发在肌肉组织内无明显的炎症排斥反应,随着HHKAT材料的降解吸收,其周围的组织反应逐渐降低.结论:研究表明人发角蛋白人工腱材料在体内能够被降解吸收,可根据不同的需要调节其降解速度,具有良好的生物相容性.

BACKGROUND: Human hair keratin artificial tendon (HHKAT) material is a new biological degradable material established in China.OBJECTIVE: To investigate the degradation of HHKAT material in vivo as well as its impact on surrounding tissues.DESIGN: A completely randomized, double-blind controlled trial was conducted.SETTING and PARTICIPANTS: The study was conducted in the Center of Experimental Animals, Kunming General Hospital of Chengdu Military Area Command of Chinese PLA. Twelve male Japanese rabbits(ordinary grade)with a body mass of (2.5 ±0. 5)kg were selected. The animals were fed with standard feed in separate cage.INTERVENTIONS: The rabbits were randomly grouped. HHKAT samples with a disposal time of 30 minutes and 15 minutes were embedded respectively in paraspinal muscle. Normal human hair was used as control group.Materials were accessed respectively at 2, 6, 12 and 24 weeks.MAIN OUTCOME MEASURES: Gross and histological observation of the material was performed in different periods.RESULTS: The degradation speed was different in HHKAT of different disposal time. The 30-minute group had the fastest degradation, which had been absorbed completely in 24 weeks; however, the 15-minute group had only partial degradation in 24 weeks; and control group had no degradation.Histological investigation showed that HHKAT and normal human hair do not evoke obvious inflammatory reactions, and the tissue reaction around gradually decreased with the degradation and absorption of HHKAT.CONCLUSION: HHKAT is a degradable and absorbable material with good biocompatibility, which the degradation speed can be adjusted according to different requirements.
