目的 探讨肿瘤坏死因子受体1(TNFR1)在肝细胞肝癌(HCC)组织中的表达及其意义.方法 应用免疫组织化学方法(SP法)检测76例HCC患者肿瘤组织和癌旁组织中TNFR1的蛋白表达,应用逆转录-聚合酶链反应(RT-PCR)检测其mRNA表达水平.结果 TNFR1在HCC中表达阳性率(39.5%),显著低于癌旁组织(89.5%,P<0.05),并且随着肿瘤分级的增加,TNFR1在癌旁组织中表达量也递增;在基因水平TNFR1 mRNA的2-△△C1值在癌旁组织和癌组织中分别为0.0730±0.0200和0.0287±0.0079(P<0.05),也同样得以验证.结论 TNFR1介导的凋亡在HCC的发展中占优势作用,TNFR1的表达增强与肝癌的发生及分化密切相关,在肝癌的发生发展中发挥重要作用.

Objective To investigate the expression of tumor necrosis factor receptor 1 (TNFR1)in hepatocellular carcinoma (HCC) and its significance.Methods The expression of TNFR1 in 76 HCC tissues and adjacent carcinoma tissues was determined with immunohistochemical methods.Meanwhile,the expression of TNFR1 mRNA was analyzed by reverse transcription-polymerase chain reaction (RT-PCR).Results The expression of TNFR1 in HCC tissues in positive rate (39.5% ) was significantly lower than the adjacent tissues (89.5%,P <0.05 ),with the increase in tumor grade,the amount of TNFR1 in cancer adjacent tissues also increased;to further substantiate observations of TNFR1 expression,the level of TNFR1 mRNA values in the adjacent tissues and cancer tissues with a mean ± standard deviation were 0.0730±0.0200 and 0.0287 ±0.0079 (P<0.05).Conclusion TNFR1 mediated apoptosis in HCC development in the dominant role,enhanced expression of TNFR1 closely related to the occurrence of liver cancer development and differentiation,and play an important role in liver cancer development.
