目的研究幽门螺杆菌(H elicobacter py lori,H py lori)简易分离培养及保存方法,构建H py lori小型菌库。方法首先采用H py lori选择培养基,以烛缸法分离培养不同来源的标本,然后以革兰染色显微镜形态检查及生化试验鉴定为H py lori,最后把鉴定为H py lori的纯培养物,分别置于布氏肉汤、50%甘油血清肉汤、50%甘油蛋清3种不同的保存液,-20°C保存,并在保存后1、6、12 d分别检测其存活率。结果培养58份临床胃镜活检标本,获13株H py lori;56名在校大学生口腔和粪便标本,获8株H py lori;共获21株H py lo i,经布氏肉汤、50%甘油血清肉汤、50%甘油蛋清3种保存液-20°C保存,21株H py lori存活率在1 d和6 d时,3者均大于90%,统计学无显著差异;12 d时,50%甘油血清肉汤存活率大于80%,布氏肉汤和50%甘油蛋清均低于50%,差异有统计学意义。结论采用烛缸法可以成功分离培养H py lori,以50%甘油血清肉汤-20°CH py lori可存活12 d以上,两者结合可以在一般实验室构建简易的过渡性H py lori小型菌库,方便H py lori菌种的使用。

Objects To study the simple method for the separation and reservation of Helicobacter pylori(H pylori) and construct a small storage of H pylori.Method Firstly,the candle jar method was used to separate samples of H pylori from different sources by H pylori selective medium,Secondarily,the Gram microscopy test and biochemical experiment were used to identify H pylori.Thirdly,the pure H pylori was reserved in three different preservation solutions(brucella broth,50% glycerol serum,50% glycerol egg white) which were 20 °C below zero.Finally,the samples were tested to find out the survival rate after 1-,6-,12-day reservation.Result Among the 21 H pylori samples,of which 13 were collected and obtained from 58 clinical specimens and the other 8 were from oral and fecal samples of 56 undergraduates,the survival rate of 21 of the samples was over 90%.That is,the survival rate showed no significant difference when they were tested after 1-and 6-day reservation.The survival rate of the samples reserved in different solutions showed significant different when tested in 12-day reservation: that of those in 50% glycerol serum was over 80%,that of those in the other solutions was less than 50%.Conclusion Candle jar method can be used as a good way to isolated H pylori and the 50% glycerol which is 20 °C below zero can reserve H pylori well for more than 12 days.It is convenient to construct a simple and small storage of H pylori and valuable in scientific research by using the bacteria from the storage.
