[目的]研究急性脑梗死患者外周血巨噬细胞移动抑制因子(macrophage migration inhibitory factor,MIF)的表达及变化水平,探讨MIF在脑梗死过程中的作用.[方法]应用ELISA法和荧光定量PCR技术,测定68例脑梗死患者急性期(1~3d)和亚急性期(10~14 d)血浆MIF的浓度和外周血单个核细胞(PBMC)MIF mRNA 的表达水平,检测同期99例健康对照者对应指标.[结果]脑梗死患者急性期血浆MIF的浓度[(52.77±25.42)ng/ml]和PBMC MIF的表达(0.42±0.37)均高于亚急性期[(39.20±17.46)ng/ml,0.27±0.29]和健康对照组[(32.66±27.92)ng/ml,0.24±0.23],差异均有显著性(P<0.05);脑梗死亚急性期血浆MIF的浓度和PRMC MIF的表达均与健康对照组无显著差异(P> 0.05).[结论]脑梗死患者血浆MIF浓度和PBM( MIF mRNA表达水平呈先增高后恢复正常的变化规律,提示MIF与脑梗死病程演变有关,动态检测血浆MIF浓度和PBMC MIF mRNA 表达水平可能对急性脑梗死的发生发展及预后有一定监测作用.

[Objective]To explore the function of peripheral blood macrophage migration inhibitory factor (MIF) during cerebral infarction by studying the expression and change of MIF in patients with cerebral infarction.[Methods]The levels of plasma MIF were measured by enzyme linked immunosorbent assay (ELISA) and the levels of peripheral blood mononuclear cells '(PBMC) MIF mRNA were measured by real-time polymerase chain reaction (RT-PCR) in 68 patients at acute stage (1~3 days),subacute stage (10~14 days).Mean time,99 healthy persons mating cerebral infarction group in both sex and ages were measured.[Results]At acute stage,the levels of plasma M1F[(52.77 ± 25.42)ng/ml]and PBMC MIF mRNA (0.42 ± 0.37)in the patients with cerebral infarction were obviously higher than those of subacute stage[(39.20 ± 17.46)ng/ml,0.27 ± 0.29]and the healthy controls[(32.66 ± 27.92)ng/ml,0.24 ± 0.23](P < 0.05).The levels of plasma MIF and PBMC MIF mRNA in the patients with cerebral infarction at subacute stage had no significant difference with those of the healthy controls (P> 0.05).[ConcIusions] The plasma level of MIF and the PBMC expression of MIF mRNA are in increased gradually,and then gradually drop down to normal level.Dynamic detections of the plasma level of MIF and the PBMC expression of MIF mRNA are useful for forecasting the development and prognosis of acute cerebral infarction.
