已分离鉴定的无色杆菌属(Achromobacter sp.)菌株MGT3对孔雀绿染料具有强脱色作用,对其产酶脱色能力进行初步研究表明,该菌株不需要孔雀绿作为底物即可产脱色酶,即脱色酶的产生不需要底物的诱导;其胞内和胞外都存在脱色能力强的脱色酶.产胞外脱色酶的条件优化结果显示培养基的组成对该菌株产胞外酶的能力有很大影响.采用单因素结合正交试验优化产酶培养基结果表明,培养基的最佳组成成分为0.5%蔗糖、2.5%尿素、0.0025%KH_2PO_4;该菌株在培养基初始pH为9、温度为35 ℃时产脱色酶效率高,对孔雀绿脱色率高.

The Achromobacter sp. MGT3 can highly decolorize malachite green. The results on its enzyme producing capability showed that the production of decoloring-enzymes did not require the induction of substrate, and the decoloring-enzymes not only existed inside the cells, but also outside the cells. The results on the exoenzyme producing conditions showed that the compositions of medium had great influence on enzyme producing capability of MGT3. Single-factor and orthogonal experiment optimizing the exoenzyme producing medium showed that the optimized compositions of medium were 0.5% sucrose, 2.5% urea and 0.0025% KH_2PO_4. The optimum pH for decolorizing malachite green was 9, and the temperature was 35 ℃.
