为快速、简单、有效的获得冠状散囊菌DNA,分别使用溶菌酶破壁法、纤维素酶破壁法、果胶酶破壁法、CTAB法、SDS法提取冠状散囊菌菌丝体和子囊孢子的基因组DNA,然后进行琼脂糖凝胶电泳、ITS序列的PCR扩增检测以及紫外分光光度计测定DNA浓度和纯度。实验结果表明:5种方法均能提取出冠状散囊菌基因组DNA,酶法中溶菌酶最好、果胶酶次之、纤维素酶较差,但三者差异不显著;SDS法明显优于CTAB法,且在五种方法中效果也是最好的,DNA浓度可达1.69mg/mL;五种方法都证实用冠状散囊菌菌丝体作为DNA提取材料优于用子囊孢子。

To choose rapid, simple and effective methods for the extraction of genomic DNA from Eurotium cristatum of threadlike hyphae and aseo-spores, the total DNA was extracted using the methods of lysozyme, eeHulase, pectinase, CTAB, and SDS, then detected by spectrophotometer, agarose gel electrophoresis. The results indicated that the five methods could be used for extracting Eurotium cristatum genomic DNA. In the three enzymes used, lysozyme was the best and peetinase was better than cellulase, but there was no obvious difference between them. SDS method was better than CTAB and the obtained DNA concentration was 1.69 mg/mL.
