以朱砂根药材的粒度、甲醇浓度、提取时间、提取次数为考察因素,采用正交设计L9(34)进行优选试验,并用反相高效液相色谱法测定提取物中岩白菜素含量。反相高效液相色谱法采用Kromasil ODS-1C18反相柱(250 mm×4.6 mm,5μm),流动相甲醇-水(体积比为25:75),流速1.0 mL.min-1,检测波长274nm,柱温25℃。最佳提取工艺为:采用超声提取法,朱砂根药材打成粉末(过40目筛)、甲醇(分析纯)为溶剂、提取次数3次、提取时间30min。

Objective: To optimize the extraction technology of bergenin from Ardisia crenata Sims.Methods : Through observing levels of the granularity,methanol concentration,extraction time and times of extraction,the optimum of methanol extraction was selected by orthogonal design L9(34).The content of bergenin in the methanol extraction was determined by RP-HPLC.RP-HPLC method was employed on a Kromasil ODS-1 C18 column(250 mm×4.6 mm,5μm) with the mobile phase of consisting of 25%(v/v) methanol.The flow rate was 1.0 mL?min-1,the detection was set at 274nm and the column temperature at 25℃.Results: The optimum extraction technology was determined as follows : Using ultrasonic extraction method with adding methanol(AR) into the powders of Ardisia crenata Sims(passed through 40 mesh sieve) and extracting three times with 30 minutes for each time.Conclusion: The optimized extraction technology is stable and feasible,and it provides theoretical basis for extraction of bergenin from Ardisia crenata Sims.
