目的:观察黄体酮对脑梗死大鼠脑组织基质金属蛋白酶9(MMP-9)和Ⅳ型胶原(collagenⅣ)表达水平的影响。方法:用线栓法制作大脑中动脉局灶性脑缺血模型(pMCAO),将成模大鼠随机分成模型组、环糊精(HBC)溶剂治疗组和黄体酮治疗组,每组设12、24、48及72h4个时间点,每个时间点6只大鼠,另选6只大鼠作为假手术组。用免疫组织化学技术检测各组大鼠脑组织MMP-9蛋白和collagenⅣ的表达情况。结果:脑梗死发生后大鼠脑组织MMP-9蛋白的表达水平均明显升高,经黄体酮治疗后其表达水平有所下降;4组间在不同时间点比较,差异均有统计学意义(P〈0.05)。collagenⅣ的表达随着缺血时间的延长而逐渐减少,经黄体酮治疗后其丢失程度有所减轻;4组间在不同时间点比较,差异均有统计学意义(P〈0.05)。结论:黄体酮可通过抑制MMP-9的表达、减少collagenⅣ的降解来减轻脑水肿。
目的:觀察黃體酮對腦梗死大鼠腦組織基質金屬蛋白酶9(MMP-9)和Ⅳ型膠原(collagenⅣ)錶達水平的影響。方法:用線栓法製作大腦中動脈跼竈性腦缺血模型(pMCAO),將成模大鼠隨機分成模型組、環糊精(HBC)溶劑治療組和黃體酮治療組,每組設12、24、48及72h4箇時間點,每箇時間點6隻大鼠,另選6隻大鼠作為假手術組。用免疫組織化學技術檢測各組大鼠腦組織MMP-9蛋白和collagenⅣ的錶達情況。結果:腦梗死髮生後大鼠腦組織MMP-9蛋白的錶達水平均明顯升高,經黃體酮治療後其錶達水平有所下降;4組間在不同時間點比較,差異均有統計學意義(P〈0.05)。collagenⅣ的錶達隨著缺血時間的延長而逐漸減少,經黃體酮治療後其丟失程度有所減輕;4組間在不同時間點比較,差異均有統計學意義(P〈0.05)。結論:黃體酮可通過抑製MMP-9的錶達、減少collagenⅣ的降解來減輕腦水腫。
목적:관찰황체동대뇌경사대서뇌조직기질금속단백매9(MMP-9)화Ⅳ형효원(collagenⅣ)표체수평적영향。방법:용선전법제작대뇌중동맥국조성뇌결혈모형(pMCAO),장성모대서수궤분성모형조、배호정(HBC)용제치료조화황체동치료조,매조설12、24、48급72h4개시간점,매개시간점6지대서,령선6지대서작위가수술조。용면역조직화학기술검측각조대서뇌조직MMP-9단백화collagenⅣ적표체정황。결과:뇌경사발생후대서뇌조직MMP-9단백적표체수평균명현승고,경황체동치료후기표체수평유소하강;4조간재불동시간점비교,차이균유통계학의의(P〈0.05)。collagenⅣ적표체수착결혈시간적연장이축점감소,경황체동치료후기주실정도유소감경;4조간재불동시간점비교,차이균유통계학의의(P〈0.05)。결론:황체동가통과억제MMP-9적표체、감소collagenⅣ적강해래감경뇌수종。
Aim:To investigate the influence of progesterone(PROG) on the expression of MMP-9 and collagen Ⅳ after focal cerebral ischemia in rats.Methods:Rat permanent middle cerebral artery occlusion (pMCAO) model was used in this experiment. All rats were randomly divided into four groups: sham operated group,ischemic group, vehicle (HBC)-treated group and PROG-treated group. Except for shamed operated group, 6 rats were killed in each group at 12,24,48 and 72 h post-occlusion, respectively. Immunohistochemistry technique was used to detect the expressions of MMP-9 and collagen Ⅳ.Results:The expression level of MMP-9 protein was higher in ischemic group and vehicle (HBC)-treated group than other two groups after stroke. There is significant difference between this four groups at all time points (P0.05). In addition, this study also revealed that the expression of MMP-9 protein was obviously down-regulated when compared with the ischemic group and the HBC treated group at all time points (P0.05). In the ischemic group the expression of collagen Ⅳ gradually decreased with time prolonging. There is significant difference between this four groups at all time points (P0.05). In the progesterone treated group, the degradation of collagenⅣ protein was obviously reduced when compared with the ischemic group at all time points (P0.05).Conclusion:Progesterone significantly reduced the expression of MMP-9 and block the down regulation of collagen Ⅳ, which is probably correlated with its effects in alleviating the degree of brain edema.
