探讨维拉帕米对大鼠缺血-再灌流视网膜Glu-IR神经元的作用。 方法:结扎双侧颈总动脉,3h后松结复流,分别于缺血前15min和再灌流前1min腹腔注 射维拉帕米(5mg/kg/次),对照组腹腔注射等量生理盐水。动物存活3d,4%多聚甲醛经心灌 注 ,视网膜切片Glu免疫组织化学反应。结果:正常大鼠视网膜内核层中有大量Glu-IR神经元 ,外核层、节细胞层部分细胞呈Glu-IR。缺血-再灌流损伤后,视网膜内核层中Glu-I R神经元较正常时显著减少,染色变淡,外核层、节细胞层未见Glu-IR(P<0.05)。在缺 血-再灌流期应用维拉帕米,视网膜中Glu-IR神经元明显增多,染色变深(P<0.05)。结 论:视网膜Glu-IR神经元对缺血-再灌流损伤敏感,维拉帕米有助于视网膜Glu-IR神经元的 存活及免疫活性的恢复。

탐토유랍파미대대서결혈-재관류시망막Glu-IR신경원적작용。 방법:결찰쌍측경총동맥,3h후송결복류,분별우결혈전15min화재관류전1min복강주 사유랍파미(5mg/kg/차),대조조복강주사등량생리염수。동물존활3d,4%다취갑철경심관 주 ,시망막절편Glu면역조직화학반응。결과:정상대서시망막내핵층중유대량Glu-IR신경원 ,외핵층、절세포층부분세포정Glu-IR。결혈-재관류손상후,시망막내핵층중Glu-I R신경원교정상시현저감소,염색변담,외핵층、절세포층미견Glu-IR(P<0.05)。재결 혈-재관류기응용유랍파미,시망막중Glu-IR신경원명현증다,염색변심(P<0.05)。결 론:시망막Glu-IR신경원대결혈-재관류손상민감,유랍파미유조우시망막Glu-IR신경원적 존활급면역활성적회복。
To investigation of verapamil on gluta mate (Glu) im munoreactivity (IR) neurons of ischemia-reperfusion retina of rat.Method s:Both left and right carotid arteries were clamped for 3h.The carotid a rteries of these animals were unclamped.Verapamil(5g/ml for verapamil-treated ra ts) or equal volume of the vehicle (for untreated animals) was peritoneal injected 1 5 minutes before 1 minute and after surger.After 3-day survival,the rats were sacr ificed.ABC method of immunohistochemistry was used to study the changes of gluta mat e in retina.Results:Glu-IR neurons distributed the inner nuclear layer,outer nuclear layers and ganglion cell of retina in t he normal rats,Afte r ischemia-reperfusion,Glu-IR neurons decreased and low stained in inner nuclea r layer,no Glu-IR neurons in outer nuclear layer and ganglion cell (P<0.05);in verapamil treated animals,Glu-IR neurons increased and high stained (P<0.05).Conclusions:Glu-IR neurons in retina may belong to the sensitive to ischemia-reperfusion,administration of verapamil may benifit the survival an d immunoreactivity of Glu-IR neurons in retina.
