目的检查疼痛刺激神经生长因子基因(NGFmRNA)在大脑的变化,探讨硫酸镁对疼痛刺激时大脑皮质NGFmRNA表达的影响。方法 32只成年Balb/c雌鼠随机分为A,B两组,A组每只腹部皮下注射10% MgSO4 500 mg*kg-1,B组注射等体积生理盐水。10 min后每只小鼠左后肢掌面皮下注射5%福尔马林40 μl。随后在3 h、6 h、12 h、24 h(每时间点4只)各时间点处死取样。另设未处理组(n=4)不经任何处理作为正常对照。在设定时间处死小鼠取大脑皮质提取总RNA,以甘油醛-3-磷酸脱氢酶(GAPDH)基因为内参照,用半定量反转录聚合酶链反应(RT-PCR)技术检测大脑皮质中NGFmRNA表达水平。结果(1)NS组Formalin注射后3 h大脑皮质NGFmRNA水平升高不明显,第6 h增幅最大,约为正常值的5倍,12h为2.5倍,24 h仍比正常值高2倍。(2)MgSO4组Formalin注射后3 h大脑NGFmRNA水平无明显变化,第6、12和24 h也比正常对照升高,但与NS组相比升高幅度显著降低(P<0.01、0.05和0.05)。结论(1)后足注射Formalin能刺激Balb/c雌鼠大脑皮质过量表达NGF mRNA。(2)Mg2+能大部分阻滞NGF mRNA在大脑皮质的表达。(3)MgSO4对中枢神经的作用与其阻滞NMDA受体活性及Ca2+内流,抑制NGF mRNA在大脑皮质的表达有关。

목적검사동통자격신경생장인자기인(NGFmRNA)재대뇌적변화,탐토류산미대동통자격시대뇌피질NGFmRNA표체적영향。방법 32지성년Balb/c자서수궤분위A,B량조,A조매지복부피하주사10% MgSO4 500 mg*kg-1,B조주사등체적생리염수。10 min후매지소서좌후지장면피하주사5%복이마림40 μl。수후재3 h、6 h、12 h、24 h(매시간점4지)각시간점처사취양。령설미처리조(n=4)불경임하처리작위정상대조。재설정시간처사소서취대뇌피질제취총RNA,이감유철-3-린산탈경매(GAPDH)기인위내삼조,용반정량반전록취합매련반응(RT-PCR)기술검측대뇌피질중NGFmRNA표체수평。결과(1)NS조Formalin주사후3 h대뇌피질NGFmRNA수평승고불명현,제6 h증폭최대,약위정상치적5배,12h위2.5배,24 h잉비정상치고2배。(2)MgSO4조Formalin주사후3 h대뇌NGFmRNA수평무명현변화,제6、12화24 h야비정상대조승고,단여NS조상비승고폭도현저강저(P<0.01、0.05화0.05)。결론(1)후족주사Formalin능자격Balb/c자서대뇌피질과량표체NGF mRNA。(2)Mg2+능대부분조체NGF mRNA재대뇌피질적표체。(3)MgSO4대중추신경적작용여기조체NMDA수체활성급Ca2+내류,억제NGF mRNA재대뇌피질적표체유관。
Objective To investigate the changes of NGF mRNA levels and explore the effects of MgSO4 on the expression of NGF mRNA in cortex of mice in formalin test. Methods Female Balb/c mice received formalin solution injection in the plantar surface of one hindpaw after 10m following MgSO4 or NS administration. Thereafter, NGF mRNA level analyses in cortex by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR)were made at 3, 6, 12 and 24 h after formalin injection(n=4,per time point). In a separate group of mice (n=4) without any treatment were sacrificed to examine normal NGF mRNA levels. Results ①The NGF mRNA expression in cortex in the mice with NS administration increased significantly at 6, 12 and 24h in comparison with that in normal control(5、2.5 and 2 fold,respectively)after formalin injection. ②The increases of NGF mRNA have been shortened significantly at 6、12 and 24 h in the group of MgSO4 compared to that in the NS (P<0.01、0.05 and 0.05,respectively) following formalin injection. Conclusion ①There is an overexpression of NGF mRNA in the cortex of female mice after formalin injection. ②MgSO4 can restrain largely the over expression of NGF mRNA in cortex of mice with formalin injection. ③We suggest that the main action of MgSO4 on nervous system, analgesia included, involve with its inhibition from the expression of NGF mRNA in central nervous system.
