目的 研究肝脏保留灌注溪黄草有效成分Nodosin对大鼠供肝缺血期的保护作用和作用机制.方法 将实验雄性Wistar大鼠随机分为6组(n=3):空白对照组(乳酸钠林格液LR组)、实验组(低浓度NL组、中浓度NM组、高浓度NH组)和阳性对照组(Co组)、阴性对照组(Zn组).实验动物经门静脉灌注实验药物,于灌注后0、1、2 h时相点取肝脏组织标本,用比色法测定肝脏SOD酶活性和MDA含量,Banff标准评价损伤的病理表现,胆红素生成试验测定HO-1酶活性.结果 NH组和Co组肝脏保留灌注2 h后,MDA含量和Banff评分明显低于LR m(p<0.05),SOD酶活性和HO-1酶活性明显高于LR组(P<0.05);Zn组肝脏保留灌注2 h后.MDA含量明显高于LR组(P<0.05),SOD酶活性和HO-1酶活性明显低于LR组(P<0.05).结论 保留灌注溪黄草有效成分Nodosin可以提高大鼠供肝抗氧化能力,减轻肝脏组织缺血期损伤,其作用可能是通过诱导肝脏组织HO-1酶表达上调来实现的.

Objective To study the mechanism of protection from ischemic injury in rat liver by reserved infusing with Nodosin. Methods Eighteen male Wistar rats were randomly assigned into 6 groups (n = 3):Control group (lacta-ted ringer's solution, LR group) , experiment groups (Nodosin -Low concentration group, Nodosin -Middle concentration group, Nodosin - High concentration group),positive control group (Copp roup),and negative control group (Znpp group). Hepatic tissues were obtained Oh, 1h and 2h after perfusion. Activity of SOD and the quantity of MDA were assessed via using colorimetric method, while the severity of pathology injury was evaluated according to Banff Criteria, and the activity of HO - 1 was assessed via using bilirubin production test. Results The quantities of MDA and Banff's scores in NH group and positive control group (Copp group) were significantly lower than that in control group (P < 0.05) at 2 h, while activities of SOD and HO - 1 were significantly higher (P<0.05). The quantity of MDA in negative control group (Znpp group) was significantly higher than that in control group (P<0.05) at 2h, while the activities of SOD and HO - 1 was significantly higher (P < 0.05). Conclusion Reserved infusion with Nodosin can improve the anti - oxidant ability of rat liver to protect from hepatic ischemic injury, which may be mediated by induction of HO - 1 expression.
