目的 建立人大肠癌(HICC)组织块裸小鼠直肠黏膜原位种植癌及转移模型.方法 人结肠癌细胞株HCT-116注射接种于裸小鼠直肠黏膜下,获得直肠种植瘤.制备该肿瘤新鲜组织块,原位种植于直肠黏膜面,在2、4、6、8、10、12周取材,进行苏木素一伊红(HE)染色及CEA免疫组织化学检测,观察种植瘤成瘤率,生长情况及转移规律.结果 直肠黏膜组织块法原位种植2周后成瘤率达90%以上.肿瘤在6~12周成指数生长.种植4周见区域淋巴结(LN)部分转移,直肠上动脉LN转移率60%、系膜LN转移率20%;6周见腹主动脉旁LN转移,转移率40%;8周见区域LN均转移充分,分别为100%、80%、80%.10~12周见肝肺转移发生,分别为20%(1/5)、40%(2/5).LN转移发生在肿瘤指数生长之前.HE及CEA免疫组织化学证实LN、肝肺有癌细胞转移浸润.结论 裸小鼠直肠黏膜植块法能成功构建HICC原位模型.此模型操作简便、制模稳定、转移率高,且生物学行为更能模拟临床HICC发生、发展演变过程.

목적 건립인대장암(HICC)조직괴라소서직장점막원위충식암급전이모형.방법 인결장암세포주HCT-116주사접충우라소서직장점막하,획득직장충식류.제비해종류신선조직괴,원위충식우직장점막면,재2、4、6、8、10、12주취재,진행소목소일이홍(HE)염색급CEA면역조직화학검측,관찰충식류성류솔,생장정황급전이규률.결과 직장점막조직괴법원위충식2주후성류솔체90%이상.종류재6~12주성지수생장.충식4주견구역림파결(LN)부분전이,직장상동맥LN전이솔60%、계막LN전이솔20%;6주견복주동맥방LN전이,전이솔40%;8주견구역LN균전이충분,분별위100%、80%、80%.10~12주견간폐전이발생,분별위20%(1/5)、40%(2/5).LN전이발생재종류지수생장지전.HE급CEA면역조직화학증실LN、간폐유암세포전이침윤.결론 라소서직장점막식괴법능성공구건HICC원위모형.차모형조작간편、제모은정、전이솔고,차생물학행위경능모의림상HICC발생、발전연변과정.
Objective To establish an orthotopic implantation and metastasis model in the mucous membrane of rectum of human intestinum crassum cancer (HICC) in nude mice. Methods Tumor cell line HCT-16 of human colon adenocarcinoma was inoculated into submucous stratum of rectum of nude mice to develop implantation tumor. Prepare the fresh tumor tussue, implant on the surface of rectal mucosa, collect the organs in 2th,4th,6th,8th, 10th, 12thweek, observing the formation rate, growth characteristics, metastasis rate of implanted tumor, detectting with H-E dyeing and immunohistochemistry stain of CEA. Results The implanted tumor rate more than 90% in 2 weeks. The tumor was exponential growth between 6 and 12 weeks. Part the region lymph node(LN) generate metastasis at 4 weeks, superior rectal artery LN metastasis rate is 60% and mesenteric LN metastasis rate is 20% ;6th week,abdominal aorta LN metastasis rate is 40% ;8th week, all the region LN metastasis sufficiently, respectively is 100% ,80%, 80%. The metastasis rate is 20% (1/5) in 10th,40% (2/5) in 12th,in the liver and lung. Metastasis generate before exponential growth, cancer cells infiltration metastasis were confirmed by H-E dyeing and immunohistochemistry stain of CEA. Conclusion This method successfully establish an orthotopic implantation and metastasis model of HICC, simply application, stability, higher metastasis and can simulate occur and develop process of disease.
