目的:初步探讨锌酞氰(ZnPc)介导的光动力学疗法(ZnPc-PDT)对小鼠Lewis肺癌细胞的杀伤作用,探索ZnPc-PDT杀伤Lewis肺癌细胞的最佳工作参数。方法:通过分光光度计测量ZnPc的吸收峰值,选取670 nm的半导体激光器作为激发光源;分别利用MTT法和流式细胞术检测光敏剂浓度、光照时间及激光功率密度对ZnPc-PDT对Lewis肺癌细胞的杀伤能力的影响。结果:ZnPc的吸收峰值为670 nm;ZnPc-PDT对肺癌细胞的杀伤作用均随光敏剂浓度、光照时间、光源功率密度的增强而增强,但增大到一定值后会趋于饱和。结论:ZnPc组分单一,与其相对应的吸收波长对组织的穿透能力较强;ZnPc介导的PDT能有效杀伤小鼠Lewis肺癌细胞,杀伤效果主要受光敏剂浓度、光照时间及光源功率密度等参数的影响。

목적:초보탐토자태청(ZnPc)개도적광동역학요법(ZnPc-PDT)대소서Lewis폐암세포적살상작용,탐색ZnPc-PDT살상Lewis폐암세포적최가공작삼수。방법:통과분광광도계측량ZnPc적흡수봉치,선취670 nm적반도체격광기작위격발광원;분별이용MTT법화류식세포술검측광민제농도、광조시간급격광공솔밀도대ZnPc-PDT대Lewis폐암세포적살상능력적영향。결과:ZnPc적흡수봉치위670 nm;ZnPc-PDT대폐암세포적살상작용균수광민제농도、광조시간、광원공솔밀도적증강이증강,단증대도일정치후회추우포화。결론:ZnPc조분단일,여기상대응적흡수파장대조직적천투능력교강;ZnPc개도적PDT능유효살상소서Lewis폐암세포,살상효과주요수광민제농도、광조시간급광원공솔밀도등삼수적영향。
Objective: To investigate the effect of ZnPc-PDT on mice Lewis lung cancer cells and explore the best parameters of ZnPc-PDT for lung cancer.Methods: We used the ultraviolet spectrophotometer to measure the absorption peak values and selected 670nm semiconductor lasers as the excitation light resources.Meanwhile,we employed MTT and FACS to observe the killing effects of ZnPC-PDT on Lewis lung cancer cells under different photosensitizers’ concentration,exposure time,and different laser power density.Results: The absorption peak values of ZnPc was 670nm,and the effects of killing lung cancer increased with the increasing of photosensitizers’ concentration,exposure time,and different light source power density,while the effects would be saturated after reaching certain value.Conclusion: ZnPc is a single components material,and its absorption wavelength has strong penetration ability to organization.ZnPc-PDT could effectively induce the death of Lewis lung cancer cells in mice.The key factors of effects of killing lung cancer are photosensitizers’ concentration,exposure time,and different laser power density.
