目的:研究葡萄糖依赖性胰岛素刺激多肽受体(Glucose-dependent insulinotmpic polypeptide receptor,GIPR)在移植胰岛血管内皮细胞的表达情况.方法:用胰管内灌注、V型胶原酶消化,Ficoll-400梯度离心纯化的方法,获取较纯胰岛后通过门静脉途径移植到糖尿病大鼠肝脏,术后每天给于免疫抑制剂他可莫斯(Taerolimus,FK506)和氯芬酸酯(Mycophenolate mofetil,MMF)灌胃.移植模型分别饲养7、14、40、60d后取材,用免疫组化方法研究GIPR在胰岛血管的表达.结果:移植胰岛内部分血管内皮细胞表达GIPR.结论:GIPR阳性内皮细胞参与了移植物血管重建;新生的胰岛血管具备肝脏血管某些特征,能够被GIP影响.

목적:연구포도당의뢰성이도소자격다태수체(Glucose-dependent insulinotmpic polypeptide receptor,GIPR)재이식이도혈관내피세포적표체정황.방법:용이관내관주、V형효원매소화,Ficoll-400제도리심순화적방법,획취교순이도후통과문정맥도경이식도당뇨병대서간장,술후매천급우면역억제제타가막사(Taerolimus,FK506)화록분산지(Mycophenolate mofetil,MMF)관위.이식모형분별사양7、14、40、60d후취재,용면역조화방법연구GIPR재이도혈관적표체.결과:이식이도내부분혈관내피세포표체GIPR.결론:GIPR양성내피세포삼여료이식물혈관중건;신생적이도혈관구비간장혈관모사특정,능구피GIP영향.
Objective: To investigate the expression of GIPR in vasculature endothelial cells in transplanted islets.Methods: Pancreas of SD rat were digested by collagenage V after the common bile ductal injection of collageenase V.Islets were purified by Ficoll discontinuous density gradient-centrifugation. Then the high-quality islets were immediately transplanted intraportally into the liver of recipient SD rats with streptozotocin-induced diabetes.The recipient rats were randomly devided into 4 groups based on time points that recipients were killed:group A(7 days)、group B(14 days)、group C(40 days),and group D(60 days).The immunosuppressive tacrolimus (FK506)and mycophenolate mofetil(MMF)were administered to recipients by intragastric administration aftertransplantation. Concentrations of blood glucose in recipient were determined with monitor.The groups were respectively killed at different time points,then islet grafts and the expression of GIPR in vasculature endothelial cells were observed morphologically. Results:GIPR was found to be expressed in parts of the microvascular endothelial cells,Conclusion: GIPR-positive endothelial cells take part in microvascular reconstruction in islet grafts.The neogenetic islet endothelial cells might acquire some common characteristics with those of liver.
