用RT-PCR方法对山东省猪瘟病毒(CSFV)流行毒株的E2基因进行克隆,用DNAstar软件对获得的CSFV及已发表的CSFV毒株E2基因核苷酸和氨基酸序列进行了比较和分析,构建了CSFV遗传发生树.结果表明:扩增的12株猪瘟流行病毒的E2基因长度均为270 bp,与预期大小一致;12株病毒株均属于1个组群中的2个亚群,其中,病毒株SDTA-1、SDTA-2属于同一个亚群,E2基因与疫苗株HCLV相比,核苷酸、氨基酸同源性分别为83.5 %、88.9 %,其余10株病毒株属于同一个亚群,E2基因与疫苗株HCLV相比,核苷酸、氨基酸同源性分别为80.5 %~83.5 %、82.2 %~84.4 %.

The major antigen coding region sequences of E2 gene of twelve recent prevalent field classical swine fever viruses(CSFV) collected from Shandong province were obtained with reverse trancriptase polymerse chain reaction (RT-PCR) and sequencing. The abovementioned sequences were analysed with DNAstar and compared with the previously published sequences of references strains. The phylogenetic tree was constructed. The results showed that all the twelve prevalent CSFV strains belonged to gene group 2 which was divided into 2 subgroups. 10 of the 12 strains were in subgroup 1 while the other 2 strains in subgroup 2. The identities of nucleotide sequence of the two subgroups was 80.5 %~83.5 %. These findings revealed that recent prevalent CSFV strains in Shandong province had been evolving towards the direction far from the C strain vaccine virus.
