由多主棒孢病菌侵染引起的棒孢霉落叶病是严重危害橡胶生产的一种世界性病害,近年来在中国也有发ⅱ生.在菌龄、细胞壁酶种类、酶作用浓度、作用时间、酶解温度、渗透压稳定剂等因素对橡胶树多主棒孢原生质体制备和再生影响分析的基础上,笔者首次建立了PEG介导的多主棒孢病菌遗传转化体系,并获得了多主棒孢病菌绿色荧光转化子.该体系中,多主棒孢病菌的原生质体再生率为19.12%,转化率为10.34个/μgDNA.
由多主棒孢病菌侵染引起的棒孢黴落葉病是嚴重危害橡膠生產的一種世界性病害,近年來在中國也有髮ⅱ生.在菌齡、細胞壁酶種類、酶作用濃度、作用時間、酶解溫度、滲透壓穩定劑等因素對橡膠樹多主棒孢原生質體製備和再生影響分析的基礎上,筆者首次建立瞭PEG介導的多主棒孢病菌遺傳轉化體繫,併穫得瞭多主棒孢病菌綠色熒光轉化子.該體繫中,多主棒孢病菌的原生質體再生率為19.12%,轉化率為10.34箇/μgDNA.

Corynespora leaf fall disease caused by Corynespora cassiicola is a serious disease of rubber plantations worldwide, and has occurred in China recently. The hypha age, types and concentrations of cell wall -degrading enzymes, time and temperatures for digestion, and osmotic pressure stabilizers were determined to observe their effects on formation and regeneration of protoplasts of C. cassiicola. The protoplast transformation system mediated by polyethylene glycol (PEG) was established, and the green fluorescent protein (GFP) transformants was obtained. The regeneration rate of protoplasts was 19.12%, and the transformation rate was 10.34 protoplasts/μgDNA.
