目的 初步探讨益肾平喘活血合剂对哮喘气道重塑大鼠血清及气道上皮炎性因子的影响.方法 SD大鼠60只,随机分为正常组,造模组,激素组和中药高、中、低剂量组各10只.按文献方法制成支气管哮喘模型.用放免法测定血清IL-2、IL-4浓度,免疫组化法测定大鼠气道TGF-β,苦味酸天狼星红染色,观察气道胶原分布和变化.结果 ①与正常组比较,中药3个组血清IL-2水平下降,IL-4水平升高均轻于造模组和激素组,与造模组比较有显著差异(P<0.01),与激素组比较无显著差异(P>0.05);中药组组间比较,高、中剂量组优于低剂量组(P<0.05).②TGF-β免疫组化造模组阳性反应最强,呈深棕色,激素组,中药3个组阳性反应稍弱,呈棕黄色至浅黄色,正常组呈淡浅黄色.③苦味酸天狼星红染色,偏振光显微镜观察,造模组支气管粘膜及上皮下Ⅰ型和Ⅲ型胶原纤维明显增多;激素组较造模组减少,中药低剂量组改变与激素组类似,中药中、高剂量组胶原纤维沉积少于激素组;正常组可见少量胶原纤维分布.结论 益肾平喘活血合剂治疗哮喘慢性持续期的作用途径,可能有一部分是通过调节IL-2、IL-4、TGF-β水平,减少气道壁胶原等来延缓支气管哮喘气道重塑的形成.

목적 초보탐토익신평천활혈합제대효천기도중소대서혈청급기도상피염성인자적영향.방법 SD대서60지,수궤분위정상조,조모조,격소조화중약고、중、저제량조각10지.안문헌방법제성지기관효천모형.용방면법측정혈청IL-2、IL-4농도,면역조화법측정대서기도TGF-β,고미산천랑성홍염색,관찰기도효원분포화변화.결과 ①여정상조비교,중약3개조혈청IL-2수평하강,IL-4수평승고균경우조모조화격소조,여조모조비교유현저차이(P<0.01),여격소조비교무현저차이(P>0.05);중약조조간비교,고、중제량조우우저제량조(P<0.05).②TGF-β면역조화조모조양성반응최강,정심종색,격소조,중약3개조양성반응초약,정종황색지천황색,정상조정담천황색.③고미산천랑성홍염색,편진광현미경관찰,조모조지기관점막급상피하Ⅰ형화Ⅲ형효원섬유명현증다;격소조교조모조감소,중약저제량조개변여격소조유사,중약중、고제량조효원섬유침적소우격소조;정상조가견소량효원섬유분포.결론 익신평천활혈합제치료효천만성지속기적작용도경,가능유일부분시통과조절IL-2、IL-4、TGF-β수평,감소기도벽효원등래연완지기관효천기도중소적형성.
Objective To investigate the effect of YiShenHuoXue decoction on serum inflammation factor of bronchus asthma airway remolding model.Methods Sixty SD rats were randomly divided into six groups: blank group,asthma group,hormone group, and high, middle and low dosage Chinese medicine groups, 10 rats in each group.The asthmatic model was established by sensitization and challenge with ovalbumin.Radioimmunoprecipitation was used to determine the contents of serum IL-2, IL-4. ELISA was used to measure the level of serum TGF-β. Picric acid sirius red dyeing was used to observe the dostritmtion and change of collagen in the rat airway. Results (1) Compared with the blank group, serum IL-2 was descreased, and IL-4 was increased in other five groups. The change of rat's serum in the three Chinese medicine groups was smaller than in the asthma group and hormone group. The levels of serum IL-2, IL-4 among Chinese medicine groups and the asthma group were different. (P<0.01). They were similar among the Chinese medicine groups and hormone group (P>0.05)The high and middle dosage groups were better than the low dosage group (P<0.05).(2) The positive reaction of serum TGF-β in the asthma group was the strongest, deep brown look. The positive reaction of serum TGF-β in the hormone group and the Chinese medicine were much lighter, brown and buff look.The blank group looked like pale buff color. (3)In picric acid sirius red dyeing and polarizable optic microscope, tapeⅠand tape Ⅲ collagen in bronchus mucous membrane and under the epidermis of the asthma group increased obviously. The change of Chinese low dosage medicine group and the hormone group were smaller, which were less than the asthma group. In the Chinese middle and high dosage medicine groups , the collagen textile fiber was less than the hormone group. There was a little collagen textile fiber in the blank group. Conclusion YiShenHuoXue decoction may control the level of rat serum IL-2 , IL-4 and TGF-β in the bronchus asthma airway remolding model, reduce the collagen in the cliff of the airway, which may slow the remodeling of bronchus in asthma rats.
