为改良拮抗姜瘟病菌的芽孢杆菌,从未发病土壤分离得到1株对茄科雷尔氏菌(Ralstonia solanacearum)等8种病原菌具有较强拮抗能力的蜡状芽孢杆菌菌株L_1.为进一步改进该菌的拮抗活性,采用紫外线(UV)、甲基磺酸乙酯(EMS)、吖啶橙等诱变因素对L_1菌株进行诱变改良.结果表明:改良后菌株的拮抗性能显著提高,其中菌株EMS-2和UL_1的拮抗蛋白浓度分别比L_1菌株提高30.38%和29.18%,其差异均达极显著水平;L_1对UV照射十分敏感,尤其在前60 s,而90 s的照射处理正突变率最高,达24.8%;吖啶橙处理L_1后,没有筛选到发生正突变的菌株;L_1在EMS处理80 min后,致死率达到90%左右,因此,为大量获取正突变菌株,应采用较低剂量的EMS(0.16 mol·mL~(-1))、长时间处理程序(60 min为宜).

To improve the resistance of Bacillus cereus to Ralstonia solanacearum, an antagonistic bacterium belonging to B. cereus, named as L_1, was isolated from the ginger-growing soil, which was resistance to R. solanacearum at some degree. Ultraviolet rays (UV), ethyl methane sulfonate (EMS) and acridine orange (AO) were used as mutagens to treat strain L_1, and then the mutants were identified and screened. The results showed that the antagonistic abilities of mutant strains were significantly enhanced compared with the original strain L_1, and the concentrations of antagonistic protein of mutant-strains EMS-2 and UL_1, were 30.38% and 29.18% higher than that of original strain L_1 separately, being significant difference between the two mutant-strains and L_1. In addition, strain L_1 was sensitive to UV especially during the period of 60 s, and up to 90 s, the utmost positive mutation rate was detected (24.8%). However, no positive mutants were founded when the strain L_1 was treated with AO. Moreover, the lethal rate of strain L_1 was up to 90% with the treatment of EMS beyond 80 min,indicating that to obtain the more number of positive mutants, the lower dosage of EMS (0.16 mol·mL~(-1)) for longer treatment time (60 min) was the possible method of mutant treatment.
