目的:探讨五味子提取物对CCl_4中毒大鼠肝细胞超微结构的保护作用.方法:Wistar大鼠25只,随机分为对照组、CCl_4中毒24 h组及10 d组、五味子24 h组及10 d组,共5组,每组5只.CCl_4中毒组大鼠用生理盐水(0.5 ml/100 g)灌胃3 d,对照组和治疗组大鼠用五味子提取液(0.5 ml/100 g)灌胃3 d后,中毒组和五味子组大鼠按0.2 ml/100 g体质量用量经腹腔一次性注射200ml/L CCl_4致毒,对照组大鼠腹腔注射同体积植物油.然后,五味子组每天按0.5 ml/100 g体质量用量给予五味子提取液灌胃,而对照组、中毒组以等量生理盐水灌胃.不同时间点处死大鼠,透射电镜下观察肝脏超微结构的改变.结果:CCl_4中毒24 h组大鼠肝组织超微结构发生改变,表现为胞质基质疏松、空泡化;肝细胞线粒体肿胀,嵴断裂或消失;内质网呈片层状;核膜皱缩、断裂,染色质边集于核膜下,CCl_4中毒10 d组肝细胞核形态仍呈明显异型性.与CCl_4中毒组大鼠相比较,五味子组大鼠肝细胞超微结构的病理性改变减轻.结论:五味子提取物对CCl_4中毒大鼠肝细胞的超微结构具有保护作用.

목적:탐토오미자제취물대CCl_4중독대서간세포초미결구적보호작용.방법:Wistar대서25지,수궤분위대조조、CCl_4중독24 h조급10 d조、오미자24 h조급10 d조,공5조,매조5지.CCl_4중독조대서용생리염수(0.5 ml/100 g)관위3 d,대조조화치료조대서용오미자제취액(0.5 ml/100 g)관위3 d후,중독조화오미자조대서안0.2 ml/100 g체질량용량경복강일차성주사200ml/L CCl_4치독,대조조대서복강주사동체적식물유.연후,오미자조매천안0.5 ml/100 g체질량용량급여오미자제취액관위,이대조조、중독조이등량생리염수관위.불동시간점처사대서,투사전경하관찰간장초미결구적개변.결과:CCl_4중독24 h조대서간조직초미결구발생개변,표현위포질기질소송、공포화;간세포선립체종창,척단렬혹소실;내질망정편층상;핵막추축、단렬,염색질변집우핵막하,CCl_4중독10 d조간세포핵형태잉정명현이형성.여CCl_4중독조대서상비교,오미자조대서간세포초미결구적병이성개변감경.결론:오미자제취물대CCl_4중독대서간세포적초미결구구유보호작용.
OBJECTIVE: To explore the protective effect of Schisandrins extract on liver ultrastructure in rats with CCl_4 poisoning. METHODS: Twenty five Wistar rots were randomly divided into the control group, the CCl_4 poisoned 24 h and 10 d groups, the Schisandrins 24 h and 10 d groups, five groups with 5 rats in each group. The rats of the CCl_4 poisoned and the Schisandrius groups were given one peritoneal injection of CCl_4(200 ml/L in vegetable oil) at a dosage of 0.2 ml/100 g body weight, the rats of the control group were given the same volume of vegetable oil. Then Schisandrins extract(0.5 ml/100 g body weight)were given orally to rats of the Schisandrins group daily,while saline was given to rats of the poisoned and the control groups. The rots were sacrificed at different time points. The morpholegic changes of the hepatoeytes were examined by transmission electron microscopy. RESULTS: The ultrastructure of hepatocytes of rats were changed in the CCl_4 poisoned 24 h group, the cytoplasm of hepatocytes was loosed and vacuolated, mitochondria were swollen, with cristae breakdown or loss, and the endoplasmic reticulum appeared lamellar. The nuclear membrane was shrunken and ruptured, and the nuclear chromatin condensed and marginated. The hepatocytic nuclei showed obvious atypia in rats of the CCl_4 10 d group. By comparing with that of the CCl_44 group,the ultrastructure of hepatocytes in rats of the schisandrins group was less severely damaged. CONCLUSION: The use of Schisandrins extract in the treatment of CCl_4 poisoned rats exerted a protective effect on liver ultrastructure.
