目的 了解绝望行为模型大鼠脑去甲肾上腺素(NE)能和5-羟色胺(5-HT)能神经元中fos蛋白(Fos)的表达情况.方法 采用不同强度强迫游泳应激造成绝望行为(BD组)模型和单纯强迫游泳模型(对照组)大鼠各10只,在其蓝斑(LC)切片上做抗Fos和抗酪胺酸羟化酶(TH)免疫组化双重染色,在中缝背核(RD)切片上做抗Fos和抗色氨酸羟化酶(TPH)免疫组化双重染色,40倍光镜F观察和计数每组LC中的TH阳性细胞数和TH/Fos 阳性细胞数、RD中的TPH阳性细胞数和5-HT/Fos阳性细胞数,并进行2组之间的比较.结果 与对照组相比,BD组LC的TH阳性细胞胞体大、染色深,TH/Fos阳性细胞数[BD组(8.9±0.7)个,对照组(3.9±0.9)个,P<0.01]明显增加,RD中出现TPH/Fos 阳性细胞[BD组(5.0±0.8)个,对照组0个,P<0.01].结论 中枢NE能神经元代谢增强导致该神经元变性件改变,可能与抑郁症的发病有关.RD巾的5-HT能神经元在正常情况下可能不参与或很少参与生理活动,只有在较强烈的应急状态下才开始被启动.

Objective To investigate the expression of fos protein (Fos) in the norepinephrinergic and serotoninergic neurons of behavioral despair rat's brain. Methods Each of 10 behavioral despair (BD) rats and simple forced swimming rats were made by using different intensity forced swimming stress. Each of immunohistochemical dual-stainning of anti-Fos/tyrosine hydroxylase (TH) or anti-Fos/tryptophan hydroxylate (TPH) were carried out on their slices of locus coemleus (LC) or nucleus raphe dorsalis (RD).The number of TH positive cells and TH/Fos positive cells on the LC and TPH positive cells and TPH/Fos positive cells on the RD observed and counted by using 40 times light microscope and compared between two groups. Results In the BD rats,TH-positive cells on the LC were swelling and staiuing were deep and the number of TH/Fos positive cells on the LC[BD group (8.9±0.7),contml group (3.9±O.9),P<0.01]and TPH/Fos-positive cells [BD group (5.0±0.8),control group O,P<0.01] increased significantly compared with the control group.Conclusion The results suggest that the degeneration of central norepinephrinergic neurons which involved increased metabolism may be relative to incidence of depression.The 5-HT neurons of RD in normal circumstances may not participate or rarely participate in physical activities and only was activated in the more strongly state of stress.
