以蝴蝶兰嫩叶、茎尖、花梗侧芽、花梗节间为外植体,进行原球茎诱导,探求蝴蝶兰组织培养的最适外植体;通过不同培养基及各种激素配比组合进行原球茎诱导、增值及试管苗生根试验,探索各阶段最优培养基配方。结果表明:茎尖和花梗腋芽是蝴蝶兰形成原球茎的较佳外植体;MS+0.5 mg/L NAA+1.0 mg/L 6-BA+100ml/L椰乳是蝴蝶兰原球茎增殖的最佳培养基,增殖系数达8.98;1/2 MS培养基最利于侧芽萌发形成丛生芽;生根最适培养基为1/2MS+1.0mg/LIBA+0.5 mg/LNAA。
以蝴蝶蘭嫩葉、莖尖、花梗側芽、花梗節間為外植體,進行原毬莖誘導,探求蝴蝶蘭組織培養的最適外植體;通過不同培養基及各種激素配比組閤進行原毬莖誘導、增值及試管苗生根試驗,探索各階段最優培養基配方。結果錶明:莖尖和花梗腋芽是蝴蝶蘭形成原毬莖的較佳外植體;MS+0.5 mg/L NAA+1.0 mg/L 6-BA+100ml/L椰乳是蝴蝶蘭原毬莖增殖的最佳培養基,增殖繫數達8.98;1/2 MS培養基最利于側芽萌髮形成叢生芽;生根最適培養基為1/2MS+1.0mg/LIBA+0.5 mg/LNAA。
이호접란눈협、경첨、화경측아、화경절간위외식체,진행원구경유도,탐구호접란조직배양적최괄외식체;통과불동배양기급각충격소배비조합진행원구경유도、증치급시관묘생근시험,탐색각계단최우배양기배방。결과표명:경첨화화경액아시호접란형성원구경적교가외식체;MS+0.5 mg/L NAA+1.0 mg/L 6-BA+100ml/L야유시호접란원구경증식적최가배양기,증식계수체8.98;1/2 MS배양기최리우측아맹발형성총생아;생근최괄배양기위1/2MS+1.0mg/LIBA+0.5 mg/LNAA。
Picking tender leaves,shoot-tips,pedicel lateral bud,pedicel internode as explants to induce original corm,to search the best explants on tissue culturing of Phalaenopsis amabilis;Different culture medium with hormone combination were screened to original corm induction,proliferation and rooting of seedling in test tube;Searching the optimal medium in different culturing stage.The result showed:the shoot-tips and pedicel lateral buds are the best explants for inducing original corm;The best culture medium of proliferation for original corm is MS+0.5 mg/L NAA+1.0 mg/L 6-BA+100ml/L coconut milk,proliferation ratio is 8.98;1/2MS culture medium is the best to induce lateral bud forming clump buds;The optimum culture medium for rooting is ?MS +1.0mg/L IBA +0.5 mg/L NAA.
