目的 观察β淀粉样前体蛋白(APP)、生长抑素(SS)在铝盐致大鼠学习记忆功能障碍模型脑内海马区表达的改变,并探讨其作用机制。方法 采用免疫组织化学方法检测β-APP、SS在大鼠脑内海马区的表达,HE染色观察实验组与对照组海马区锥体细胞层神经元形态结构。结果 实验组大鼠学习记忆功能较对照组明显降低(P<0.01);实验组海马区β-APP表达较对照组明显增加(P<0.01);SS免疫反应性神经元数目却显著减少(P<0.01);β-APP阳性度与SS阳性度之间存在等级负相关(P<0.05),实验组与对照组海马区神经元形态结构均基本正常,数目无明显差异。结论 铝盐致大鼠学习记忆障碍的病理机制可能与脑内β-APP、SS 表达的改变相关。

목적 관찰β정분양전체단백(APP)、생장억소(SS)재려염치대서학습기억공능장애모형뇌내해마구표체적개변,병탐토기작용궤제。방법 채용면역조직화학방법검측β-APP、SS재대서뇌내해마구적표체,HE염색관찰실험조여대조조해마구추체세포층신경원형태결구。결과 실험조대서학습기억공능교대조조명현강저(P<0.01);실험조해마구β-APP표체교대조조명현증가(P<0.01);SS면역반응성신경원수목각현저감소(P<0.01);β-APP양성도여SS양성도지간존재등급부상관(P<0.05),실험조여대조조해마구신경원형태결구균기본정상,수목무명현차이。결론 려염치대서학습기억장애적병리궤제가능여뇌내β-APP、SS 표체적개변상관。
Objective To observe the expression of β-APP and SS in the hippocampus of the rat mode-ls with the deficits of learning and memory function, and investigate its mechanism.Methods Using immunohistochemistry (IHC), we detected the expressions of β-amyloid precursorprotein(β-APP) and somatostatin(SS) in the hippocampus of the rat brains,using HE staining method, we observed the shapes and structures of pyramidal cell layer neurons in hippocampus of the rats in the experimental group and the controls.Results The learning and memory functions were lower significantly in the experimental group than in the controls (P<0.01);The expression of β-APP in hippocampus of experimental group was significantly higher than the controls (P<0.01);but the number of SS immunoreactive positive neurons decreased sigificantly (P<0.01); There was negative relation between the positive degrees of β-APP and SS in the experimental group (P<0.05).The structures of neurons in hippocampus of the experimental group and the controls were fundamental normal,there was no significant difference between the numbers of their neurons.Conclusion The pathogenesis of aluminum slat resulted in the deficits of learning and memory functions in rats,it might be related to the changes of the expressions of β-APP and SS in brain.
