目的:研究四味镇痛中药对内腓肽降解酶的作用.方法:从大鼠的肾脏制备含有内腓肽降解酶的物质,建立内腓肽降解酶活性检测模型;并用此模型观察四味镇痛中药提取物对内腓肽降解酶特别是对中性内肽酶活性(NEP24.11)的抑制作用.结果:(1)钩藤和羌活的水提物显示很强的NEP24.11酶抑制作用;(2)无论是有机提取物还是水提物,延胡索和川芎对NEP24.11的抑制作用都弱于钩藤和羌活;(3)钩藤和羌活不仅是氨肽酶和中性内肽酶的双重抑制剂,而且是氨肽酶、中性内肽酶和血管紧张素转化酶的三重抑制剂.结论:四味镇痛中药具有不同的镇痛机制,其中,钩藤和羌活通过抑制氨肽酶和中性内肽酶而发挥镇痛作用.

목적:연구사미진통중약대내비태강해매적작용.방법:종대서적신장제비함유내비태강해매적물질,건립내비태강해매활성검측모형;병용차모형관찰사미진통중약제취물대내비태강해매특별시대중성내태매활성(NEP24.11)적억제작용.결과:(1)구등화강활적수제물현시흔강적NEP24.11매억제작용;(2)무론시유궤제취물환시수제물,연호색화천궁대NEP24.11적억제작용도약우구등화강활;(3)구등화강활불부시안태매화중성내태매적쌍중억제제,이차시안태매、중성내태매화혈관긴장소전화매적삼중억제제.결론:사미진통중약구유불동적진통궤제,기중,구등화강활통과억제안태매화중성내태매이발휘진통작용.
AIM: To study the effects of four Chinese herbal extracts on enkephalin-hydrolysing enzymes.METHODS: The crude microsomal fraction containing enkephalin-hydrolysing enzymes was prepared from therat kidney and the enkephalin-hydrolysing enzymes assay was established. Then the inhibitory effects of fourChinese herbal extracts on enkephalin-hydrolysing enzymes were observed in vitro. RESULTS: The experi-ments showed that the aqueous extract of Gou teng(Uncaaria rhynchophylia Jacks. )and Qiang huo(Angelicagrosserrata Maxim. )have significant inhibitory effects on NEP but results with organic solvent extracts werenot significant. Both aqueous and organic extracts of Yan husu(Corydalis ambiguach. Etschl. )and Chuan xiong(Conioselinum unvittatum Turcz)showed less inhibitory activity than that of Gou teng and Qiang huo. The ex-periments showed that Gou teng water extract(Gwext)and Qianghuo water extract( Qwext)were not only dualinhibitors but also triple inhibitors of NEP/APN/ACE. CONCLUSION: This suggested that the four herbs mayhave different bases for their analgesic activity. Gou teng and Qiang huo have the analgesic activity by way ofinhibiting APN and NEP24.11.
