[目的]筛选最适的长春花ISSR-PCR 反应体系.[方法]采用正交试验方法,对影响长春花ISSR-PCR反应体系的引物浓度、dNTP浓度、Mg~(2+)浓度、Taq DNA聚合酶浓度和模板DNA浓度5个因素在4个水平上进行正交试验,建立适合长春花ISSR-PCR的反应体系.[结果]长春花ISSR-PCR 反应体系的最佳条件:引物浓度0.50 μmol/L,dNTP浓度0.10 mmol/L,Mg~(2+)浓度 3.00 mmol/L,Taq 酶浓度0.75 U/25 μl,DNA模板浓度350 ng/25 μl.采用该反应体系筛选出12对适合于长春花ISSR-PCR反应的引物.[结论]利用优化系统进行长春花的ISSR-PCR反应,可获得稳定性高、重复性好、背景清晰的电泳结果.

[Objective] The optimal ISSR-PCR reaction system of madagascar periwinkle(Catharanthus roseus) was screened.[Method] The suitable ISSR-PCR reaction system of madagascar periwinkle was established through the study on the effect of the concentration of primers,dNTP concentration,Mg~(2+) concentration,Taq DNA polymerase and template DNA concentration on the ISSR-PCR reaction system with the orthogonal test method with four levels.[Result] The best condition of the ISSR-PCR reaction system was follows:the concentration of primer,0.50 μmol/L; dNTP,0.10 mmol/L; Mg~(2+),3.00 mmol/L; Taq enzyme,0.75 U/25 μl and DNA template,350 ng/25 μl.12 pairs of suitable ISSR-PCR reaction primers could be selected with this reaction system.[Conclusion] With the optimized ISSR-PCR reaction system of madagascar periwinkle,the good results with high stability,reproducibility and clear background of electrophoresis could be obtained.
