通过对分别来自四川、湖北、浙江、上海的麦冬(Ophiopogon japonicus(L.f.)Ker-Gawl.)的13个居群和两个沿阶草种(Ophiopogon bodinieri Lévl和Ophiopogon bockianus Diels)的RAPD分析,研究麦冬的道地性.所用麦冬和两个近缘种同属沿阶草属.从40条随机引物中筛选到8条引物用作RAPD分析.通过分析获得了504条RAPD带,其中多态性占89.4%.根据距离系数构建了13个麦冬居群和两个沿阶草种的非加权算术平均(UPGMA)树.最大的遗传距离是在麦冬居群和两个沿阶草种间.在麦冬种中,采自四川大学的大叶型与其他居群距离最大.在不同地理分布和不同叶型的麦冬之间具有高度的遗传多样性.证明RAPD是一个能有效区分麦冬与其他同属物种以及麦冬道地性分析的方法.

To identify Ophiopogon japonicus (L. f. ) Ker-Gawl. as geo-herbal, 13 populations of O. japonicus (L. f. ) Ker-Gawl. and two other species, Ophiopogon bodinieri L évl and Ophiopogon bockianus Diels, were investigated using the random amplified polymorphic DNA (RAPD) method. All natural populations of O. japonicus (L. f. ) Ker-Gawl. from Sichuan, Hubei, Zhejiang, Shanghai, and two other species belong to the same genus Ophiopogon. Eight RAPD primers selected from 40 random decamer oligonucleotides were used for the analysis. Five hundred and four RAPD bands were generated by the 8 selected primers and the percentage of polymorphic bands was 89.4% on average. The 13 populations of O. japonicus (L. f. ) Ker-Gawl.and two other species were clustered in unweighted pair group method with arithmetic average (UPGMA) tree. The largest distance was found between O. bockianus Diels, O. bodinieri Lévl.and the populations of O. japonicus (L. f. ) Ker-Gawl.. Among the O. japonicus (L. f. ) Ker-Gawl., the largest distance was found between big leaf type population collected from Sichuan University and the other populations. A high level of genetic variation occurred among the different clime and leaf-type populations. The authors supported that RAPD analysis distinguish O. japonicus (L. f. ) Ker-Gawl. from other species of genus Ophiopogon and identify the population of O.japonicus (L. f. ) Ker-Gawl. used as geo-herbal.
