Zhennao-Ning capsule%Migraine%Microcirculatory impairment
目的 观察镇脑宁胶囊对动物脑微循环的影响.方法 采用结扎右侧颈总动脉法建立小鼠脑缺血模型,描记造模前5 min、造模后10、20、30 min小鼠脑血流量,计算血流量下降率;采用静脉注射高分子右旋糖苷法致大鼠脑微循环障碍,记录出现微循环障碍的时间,以及注射20 min后同一视野的血管流速、血液流态以及网点数.结果 结扎右侧颈总动脉后,脑缺血小鼠于10、20、30 min大脑皮层血流量明显下降,分别为(71±13)%、(71±15)%和(74±16)%(P<0.01);与模型组比较,镇脑宁胶囊1500 mg/kg和750 mg/kg给药后,在结扎后10、20、30 min内,小鼠大脑皮层血流量均有明显增加(56%、51%、50%和42%、49%、51%,P<0.01).注射高分子右旋糖苷后,微循环障碍模型组大鼠的血液流速差为( 572±177) μm/s,出现微循环障碍时间为(232±70)s;镇脑宁胶囊600 mg/kg和300 mg/kg给药后均能使血液流态改变减轻(P<0.01),血液流速分别增加26%,36%,出现微循环障碍的时间分别延长53%,34% (P<0.01或P<0.05).结论 镇脑宁胶囊通过改善动物脑微循环治疗偏头痛.

목적 관찰진뇌저효낭대동물뇌미순배적영향.방법 채용결찰우측경총동맥법건립소서뇌결혈모형,묘기조모전5 min、조모후10、20、30 min소서뇌혈류량,계산혈류량하강솔;채용정맥주사고분자우선당감법치대서뇌미순배장애,기록출현미순배장애적시간,이급주사20 min후동일시야적혈관류속、혈액류태이급망점수.결과 결찰우측경총동맥후,뇌결혈소서우10、20、30 min대뇌피층혈류량명현하강,분별위(71±13)%、(71±15)%화(74±16)%(P<0.01);여모형조비교,진뇌저효낭1500 mg/kg화750 mg/kg급약후,재결찰후10、20、30 min내,소서대뇌피층혈류량균유명현증가(56%、51%、50%화42%、49%、51%,P<0.01).주사고분자우선당감후,미순배장애모형조대서적혈액류속차위( 572±177) μm/s,출현미순배장애시간위(232±70)s;진뇌저효낭600 mg/kg화300 mg/kg급약후균능사혈액류태개변감경(P<0.01),혈액류속분별증가26%,36%,출현미순배장애적시간분별연장53%,34% (P<0.01혹P<0.05).결론 진뇌저효낭통과개선동물뇌미순배치료편두통.
Objective To observe the effects of Zhennao-Ning capsule on cerebral microcirculation.Methods Cerebral hypoperfusion model of mices was induced by right common carotid arteries ligation method,The cerebral blood flow for 5min was recorded under normal circumstances comand for 30min when modeled,and then calculate the rate of blood flow changes.Microcirculatory impairment model of rats was induced by intravenous injection of high molecular dextran(HMD).The time of the emergence of microcirculatory impairment was recorded.Vascular flow,blood flow pattern and the number of outlets in the same range were recorded.Results The blood flow was significant decrease in the cerebral hypoperfusion model mice,when compared with the control mice.Zhennao-Ning capsules at the dosage of 1500 mg/kg、750 mg/kg treatment could improve the cerebral blood flow of rats at 10,20,and 30 min after modeled (56%,51%,50% and 42%,49%,51%,respectively,P<0.01).Zhennao-Ning capsules at the dosage of 600 mg/kg,300 mg/kg could significantly reduce the change of blood flow pattern(P<0.01),improve vascular flow rate (26%,36%,P<0.05 or P<0.01),extend the time of the emergence in the microcirculatory impairment model rat (53%,34%,P<0.01).Conclusion Zhcnnao-Ning capsules could improve microcirculatory impairment and partly cure the clinical migraine disease.
