目的:观察热矿泥浴对大鼠佐剂性关节炎( AIA)的治疗效果及作用机制. 方法: Wistar大鼠按 AIA造模后,随机分为 3组,分别给予热矿泥浴、普通黄泥浴共 19 d,空白对照组则不予处理.连续观察关节炎症积分,第 30天取标本,检测血清肿瘤坏死因子( TNF α)含量,关节的组织病理学改变,原位杂交法观察基质金属蛋白酶-1( MMP-1)和细胞间黏附分子( ICAM-1) mRNA在关节滑膜和软骨中的表达. 结果:热矿泥浴能显著降低 AIA的关节炎症程度,包括炎症积分,以及组织切片的滑膜细胞增生和软骨侵袭.热矿泥浴还能显著降低血液中的 TNF α含量,抑制滑膜和软骨细胞的 MMP-1 mRNA以及软骨细胞 ICAM-1 mRNA的表达.普通黄泥浴也能抑制软骨细胞的 ICAM-1 mRNA的表达. 结论:热矿泥浴能显著抑制大鼠 AIA的关节炎症,其机制与抑制 TNF α的产生以及 MMP-1和 ICAM-1在滑膜和软骨中的表达有关,且这种作用不单纯是泥浴的物理作用,可能主要是热矿泥浴的化学和生物学作用所致.

목적:관찰열광니욕대대서좌제성관절염( AIA)적치료효과급작용궤제. 방법: Wistar대서안 AIA조모후,수궤분위 3조,분별급여열광니욕、보통황니욕공 19 d,공백대조조칙불여처리.련속관찰관절염증적분,제 30천취표본,검측혈청종류배사인자( TNF α)함량,관절적조직병이학개변,원위잡교법관찰기질금속단백매-1( MMP-1)화세포간점부분자( ICAM-1) mRNA재관절활막화연골중적표체. 결과:열광니욕능현저강저 AIA적관절염증정도,포괄염증적분,이급조직절편적활막세포증생화연골침습.열광니욕환능현저강저혈액중적 TNF α함량,억제활막화연골세포적 MMP-1 mRNA이급연골세포 ICAM-1 mRNA적표체.보통황니욕야능억제연골세포적 ICAM-1 mRNA적표체. 결론:열광니욕능현저억제대서 AIA적관절염증,기궤제여억제 TNF α적산생이급 MMP-1화 ICAM-1재활막화연골중적표체유관,차저충작용불단순시니욕적물리작용,가능주요시열광니욕적화학화생물학작용소치.
AIM:To observe the therapeutic effect and mechanism of thermal mineral mud-bath(TMM) against adjuvant-induced arthritis(AIA) in rats. METHODS:Wistar rats with AIA were divided into three group randomly, treated with TMM or loess mud-bath(LM) for 19 days,respectively.The control group was treated with nothing.The arthritic scores were observed constantly until the 30th day,then the specimen of blood and joints was obtained.The serum tumor necrosis factor α (TNF α )were measured by ELISA, the histopathology of the inflamed ankle were observed,and the expression of matrix metalloproteinase 1(MMP-1) mRNA and intercellular adhesion molecule 1(ICAM-1) mRNA in the synovium and cartilage were detected using in situ hybridization. RESULTS:TMM significantly decreased the severity of arthritis,including arthritic scores,synovium cells proliferation and cartilage erosion in histopathology.Besides,TMM clearly deduced the content of serum TNF α and suppressed the expressions of MMP-1 mRNA in synovium cells and chondrocytes as well as ICAM-1 mRNA in chondrocytes.Meanwhile,LM also suppressed the expression of ICAM-1 mRNA in chondrocytes. CONCLUSION:TMM significantly inhibited adjuvant-induced arthritis in rats, with suppression of serum TNF α production and the MMP-1 and ICAM-1 mRNA expression in synovium and cartilage. This therapeutic effect attributed to the colligation of physical chemical and biological effects from the thermal mineral mud-bath.
