目的 观察连花清瘟胶囊对吸烟大鼠Clara细胞和氧化应激的影响.方法 将24只雄性Wistar大鼠随机分为正常组、吸烟组、连花清瘟胶囊小剂量和大剂量治疗组,每组6只.电镜观察Clara细胞超微结构改变,免疫组化方法检测肺组织CC16蛋白表达,荧光测定法测定血浆和肺组织还原型谷胱甘肽(GSH)及氧化型谷胱甘肽(GSSG)含量.结果 吸烟3周后,吸烟组大鼠Clara细胞胞浆内分泌颗粒减少,细支气管CC16阳性细胞、血浆GSH浓度及GSH/GSSG比值降低(P值均<0.05).与吸烟组相比,连花清瘟胶囊小剂量治疗组Clara细胞分泌颗粒增加,呼吸性细支气管CC16阳性细胞比例和血浆GSH浓度增加(P<0.05);大剂量治疗组呼吸性细支气管CC16阳性细胞比例增加(P<0.05),血浆GSH浓度无差异.相关性分析表明,Clara细胞阳性指数评分与血浆GSH浓度呈正相关(r=0.617,P<0.01).结论 小剂量连花清瘟胶囊可减轻吸烟所致大鼠Clara细胞结构及功能的损害和全身氧化应激反应.

목적 관찰련화청온효낭대흡연대서Clara세포화양화응격적영향.방법 장24지웅성Wistar대서수궤분위정상조、흡연조、련화청온효낭소제량화대제량치료조,매조6지.전경관찰Clara세포초미결구개변,면역조화방법검측폐조직CC16단백표체,형광측정법측정혈장화폐조직환원형곡광감태(GSH)급양화형곡광감태(GSSG)함량.결과 흡연3주후,흡연조대서Clara세포포장내분비과립감소,세지기관CC16양성세포、혈장GSH농도급GSH/GSSG비치강저(P치균<0.05).여흡연조상비,련화청온효낭소제량치료조Clara세포분비과립증가,호흡성세지기관CC16양성세포비례화혈장GSH농도증가(P<0.05);대제량치료조호흡성세지기관CC16양성세포비례증가(P<0.05),혈장GSH농도무차이.상관성분석표명,Clara세포양성지수평분여혈장GSH농도정정상관(r=0.617,P<0.01).결론 소제량련화청온효낭가감경흡연소치대서Clara세포결구급공능적손해화전신양화응격반응.
Objective To investigate the effects of Lianhuaqingwen capsule on Clara cell and oxidative stress in smoking rats.Methods Twenty-four male Wistar rats were randomly divided into four groups:normal group,smoke exposure group,and two groups of smoking rats treated with low dose and high dose of Lianhuaqingwen capsule.The ultrastructure of Clara cell was observed by electron microscope.The expression of Clara cell secretary protein(CC16)was determined by immunohistochemistry.The levels of reduced glutathione(GSH)and oxidized glutathione(GSSG)in plasma and lung tissue were measured by iluorimetry.Results After smoke exposure for three weeks,the proportion of CC16-positive cells in bronchioles,immunostaining intensity of Clara cells,GSH level and the ratio of GSH/GSSH in plasma of smoke exposure group decreased(all P<0.05).With the use of Lianhuaqingwen capsule,the proportion of CC16-positive cells in respiratory bronchioles of low dose and high dose treatment groups and the GSH level in plasma of low dose treatment group were higher than those of smoke exposure group(all P<0.05).Correlation analysis showed that the immunostaining intensity of Clara cell was positively associated with GSH level in plasma(r=0.617,P<0.01).Conclusions Low dose of Lianhuaqingwen capsule may alleviate Clara cell damage and systemic oxidative stress of rats induced by smoking.
