目的 观察血小板活化的标志-- P-选择素在原发性高血压伴腔隙性脑梗死患者发病机制中的作用. 方法利用 FITC标记的抗 CD42a单抗(鼠 IgG1) ,PE标记的抗 CD62P单抗(鼠 IgG1) ,CY标记的抗 CD45单抗(鼠 IgG1) ,FITC标记的鼠 IgG1同种型对照免疫球蛋白 ,PE标记的鼠 IgG1同种型对照免疫球蛋白,上述单抗分别标记全血中血小板及白细胞的相关抗原;同时利用流式细胞仪,采取全血三色法流式细胞术方法,检测全血血小板上 P-选择素阳性表达:全血标本中分别加入 3种荧光标记的单克隆抗体 FITC -CD42a, PE-62P及 CY- CD45,室温避光反应 20 min, 10g/L多聚甲醛固定 10 min,用 PBS洗 2次,上机分析. 结果原发性高血压伴腔隙性脑梗死患者血小板 P-选择素阳性表达 [( 23.0± 4.0)% ]明显高于原发性高血压无腔隙性脑梗死患者 [( 16.9± 1.8%) ]及正常人组 [(5.2± 0.7)% ],三者之间相比差异有显著性意义( t=137.4,28.4, P< 0.05). 结论 P-选择素在原发性高血压伴腔隙性脑梗死发病中起重要作用, P-选择素与血管渐进损伤有关.
目的 觀察血小闆活化的標誌-- P-選擇素在原髮性高血壓伴腔隙性腦梗死患者髮病機製中的作用. 方法利用 FITC標記的抗 CD42a單抗(鼠 IgG1) ,PE標記的抗 CD62P單抗(鼠 IgG1) ,CY標記的抗 CD45單抗(鼠 IgG1) ,FITC標記的鼠 IgG1同種型對照免疫毬蛋白 ,PE標記的鼠 IgG1同種型對照免疫毬蛋白,上述單抗分彆標記全血中血小闆及白細胞的相關抗原;同時利用流式細胞儀,採取全血三色法流式細胞術方法,檢測全血血小闆上 P-選擇素暘性錶達:全血標本中分彆加入 3種熒光標記的單剋隆抗體 FITC -CD42a, PE-62P及 CY- CD45,室溫避光反應 20 min, 10g/L多聚甲醛固定 10 min,用 PBS洗 2次,上機分析. 結果原髮性高血壓伴腔隙性腦梗死患者血小闆 P-選擇素暘性錶達 [( 23.0± 4.0)% ]明顯高于原髮性高血壓無腔隙性腦梗死患者 [( 16.9± 1.8%) ]及正常人組 [(5.2± 0.7)% ],三者之間相比差異有顯著性意義( t=137.4,28.4, P< 0.05). 結論 P-選擇素在原髮性高血壓伴腔隙性腦梗死髮病中起重要作用, P-選擇素與血管漸進損傷有關.
목적 관찰혈소판활화적표지-- P-선택소재원발성고혈압반강극성뇌경사환자발병궤제중적작용. 방법이용 FITC표기적항 CD42a단항(서 IgG1) ,PE표기적항 CD62P단항(서 IgG1) ,CY표기적항 CD45단항(서 IgG1) ,FITC표기적서 IgG1동충형대조면역구단백 ,PE표기적서 IgG1동충형대조면역구단백,상술단항분별표기전혈중혈소판급백세포적상관항원;동시이용류식세포의,채취전혈삼색법류식세포술방법,검측전혈혈소판상 P-선택소양성표체:전혈표본중분별가입 3충형광표기적단극륭항체 FITC -CD42a, PE-62P급 CY- CD45,실온피광반응 20 min, 10g/L다취갑철고정 10 min,용 PBS세 2차,상궤분석. 결과원발성고혈압반강극성뇌경사환자혈소판 P-선택소양성표체 [( 23.0± 4.0)% ]명현고우원발성고혈압무강극성뇌경사환자 [( 16.9± 1.8%) ]급정상인조 [(5.2± 0.7)% ],삼자지간상비차이유현저성의의( t=137.4,28.4, P< 0.05). 결론 P-선택소재원발성고혈압반강극성뇌경사발병중기중요작용, P-선택소여혈관점진손상유관.
AIM:To detect the effect of P-selectin,the marker of platelet activation in primary hypertension associated with lacunar infarction. METHODS:FITC-labeled antiCD42a monoclonal antibody(mice IgG1),PE labeled antiCD62P monoclonal antibody(mice IgG1),CY labeled antiCD45 monoclonal antibody(mice IgG1),FITC labeled mice IgG1 isotype matched immune globulin,PE labeled mice IgG contrast immunoglobulin were used to label corresponding antigens of platelets and leukocytes in whole blood.At the same time, P-selectin on the platelet was detected by the method of three-color cytometery in whole blood;Three monoclonal antibodies with fluorescence labeled, FITC-CD42a, PE -62P and CY-CD45 were added in whole blood specimen and incubated for 20 min without light and under room temperature,over night.Then specimen was fixed with citromint for 10 min and washed with PBS twice. RESULTS:The expression of P selectin in hypertension with lacunar infarction patients[(23.0± 4.0)% ] was higher than that in hypertension without lacunar infarction patients[(16.9± 1.8)% ],as well as higher than that of in normal persons[(5.2± 0.7)% ,t=137.4,28.4,P< 0.05]. CONCLUSION:P-selectin plays an important role in primary hypertension associated with lacunar infarction which is related to the progressive injury of vessels.
