目的 探讨白介素一-3(IL-13)对正常人表皮成纤维细胞(NHDF)产生炎症因子的影响.方法 使用不同浓度IL-13(0.1、1、10 ng/m1)刺激体外培养的NHDF,应用ELISA方法榆测IL-13刺激NHDF 24h后,白介素-6(IL-6),白介素-8(IL-8),巨噬细胞趋化蛋白-1(MCP-1)和嗜酸性粒细胞趋化因子(Eotaxin)的蛋白表达水平.结果 0.1 ng/ml IL-13刺激下,IL-6、MCP-1和Eotaxin的蛋白表达水平分别为(727.33±39.88)pg/ml、(414.67±13.01)pg/ml、(124.67±8.67)pg/ml;1 ng/ml IL-13刺激下,分别为(1222.50±49.95)pg/ml、(705.33±25.15)pg/ml、(205.33±16.59)pg/ml;10 as/ml IL-13刺激下,分别为(1502.83±5.31)pg/ml、(1329.17±56.08)pg/ml、(457.50±15.32)pg/ml,均明最高于对照组[(283.50±15.86)pg/ml、(205.17±3.76)pg/ml、(13.50±5.01)pg/ml,P<0.05或0.01],并具有浓度依赖性.IL-8蛋白表达水平与对照组无显著性差异(P>0.05).结论 IL-13可刺激NHDF产生IL-6、MCP-1和Eotaxin,这些炎症因子的大量表达可能参与了IL-13在皮肤组织纤维化过程巾的病理性作用.

목적 탐토백개소일-3(IL-13)대정상인표피성섬유세포(NHDF)산생염증인자적영향.방법 사용불동농도IL-13(0.1、1、10 ng/m1)자격체외배양적NHDF,응용ELISA방법유측IL-13자격NHDF 24h후,백개소-6(IL-6),백개소-8(IL-8),거서세포추화단백-1(MCP-1)화기산성립세포추화인자(Eotaxin)적단백표체수평.결과 0.1 ng/ml IL-13자격하,IL-6、MCP-1화Eotaxin적단백표체수평분별위(727.33±39.88)pg/ml、(414.67±13.01)pg/ml、(124.67±8.67)pg/ml;1 ng/ml IL-13자격하,분별위(1222.50±49.95)pg/ml、(705.33±25.15)pg/ml、(205.33±16.59)pg/ml;10 as/ml IL-13자격하,분별위(1502.83±5.31)pg/ml、(1329.17±56.08)pg/ml、(457.50±15.32)pg/ml,균명최고우대조조[(283.50±15.86)pg/ml、(205.17±3.76)pg/ml、(13.50±5.01)pg/ml,P<0.05혹0.01],병구유농도의뢰성.IL-8단백표체수평여대조조무현저성차이(P>0.05).결론 IL-13가자격NHDF산생IL-6、MCP-1화Eotaxin,저사염증인자적대량표체가능삼여료IL-13재피부조직섬유화과정건적병이성작용.
Objective To investigate the effect of interleukin-13 (IL-13) on inflammation rehted cytokines production in normal human dermal fibroblasts (NHDF). Methods In vitro cultured NHDF with different concentrations of IL-13(0.1ng/ml,1ng/ml,10ng/ml)for 24 hours, the ELISA was applied to measure the protein level of interleukin-6 (IL-6),interleukin-8 (IL-8), monocyte-chemoattracting protein-1 (MCP-1) and Eotaxin in the supernatant. Results IL-6,MCP-1 and Eotaxin protein productions were significantly and dose dependently increased in IL-13 treated NHDF (P <0.05), while IL-8 production was not significantly increased (P>0.05). Conclusions IL-13 can stimulate NHDF producing more IL-6, MCP-1 and Eotaxin. These inflammation related eytokines may be involved in the pathogenesis of skin fibrosis.
