为研究水域环境中双酚A(BPA)污染对两栖动物精巢中芳香化酶(P450arom)和雌激素受体(ER)的影响,将中国林蛙(Rana chensinensis)雄性成体分别在精原细胞增殖期、精母细胞成熟分裂期和精子形成期暴露于浓度为5×10-7、5×10-8mol/L BPA的水体中30 d,并设5×10-9、5×10-10mol/L雌二醇(E2)阳性对照和空白对照.用免疫组织化学方法检测精巢中P450arom和ER的表达.结果表明,P450arom和ER在林蛙精巢Leydig细胞和Sertoli细胞中均存在不同程度的表达.在精巢的3个发育时期,BPA均可导致P450arom和ER表达上调;但BPA对P450arom的诱导作用在3个时期之间的差异不显著,而对ER的诱导作用在3个时期间存在显著差异,其中在精子形成期的作用最强.提示BPA对林蛙精巢功能的影响不仅通过干扰性激素合成途径中的P450arom,同时还高调ER的表达.

In order to investigate the effects of bisphenol A(BPA) in water environment on aromatase(P450arom)and estrogen receptor(ER)in the testis of amphibian,adult male frogs Rana chensinensis were exposed to 5×10-7,5×10-8 mol/L BPA for 30 d during spermatogonium proliferation,spermatocyte meiosis and spermatoleosis stages of the testis,respectively.In addition,5×10-9,5×10-10 mol/L 17 β-esradiol(E2) were chosen as positive control.The expressios of P450 arom and ER in testis were detected by immunohistochemistry.The results indicated that P450 arom and ER in Leydig and Sertoli cells of the testis were localized with varying degrees of expression.BPA up-regulates the expression of P450 arom and ER in testis of R.chensinensis during the three stages.There was no significant difference in the inductive effects of bisphenol A on P450 arom,while ER was significant different during the three stages,and the strongest effects were showed at the spermatoleosis stage.These results suggest that the effects of bisphenol A on testicular function were carried out by interfering with P450 arom in sex hormone synthesis pathway,and up-regulating the expression of ER.
