为了解硬粒小麦和野生二粒小麦的高分子量谷蛋白亚基(HMW-GS)组成及优质亚基的分布特点,并对其多样性进行分析,利用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)对32份硬粒小麦和75份野生二粒小麦共107份材料的HMW-GS组成进行了分析.结果表明,Glu-1位点共有27种等位基因,其中Glu-A1位点4种,Glu-B1位点23种;亚基null和6+8在各自的位点上出现频率最高,分别达到38.84%和16.82%;其亚基组成类型共有47种,主要为1/6+8,频率达7.48%;同时筛选出33份含有1、2~*、13+16、14+15、17+18等优质亚基的材料,可作为优质基因源.多样性分析结果表明,在Glu-A1和Glu-B1位点以及HMW-GS组成上,野生二粒小麦的多样性都大于硬粒小麦.另外,在材料GW-2、GW-4中发现未命名的新亚基.

In order to know the genetic diversity of high molecular weight gluten, 32 durum wheat and 75 wild emmer wheat were analyzed by means of SDS-PAGE. The result indicated that there were 27 alleles on Glu-1 loci, including 4 alleles on Glu-A1 loci, 23 alleles on Glu-B1 loci;And null, 6+8 subunits occurred with highest frequency on each loci, reaching 38.84% and 16.82%, respectively. There were 47 types of subunit compositions and the predominant was 1/6+8 with a frequency of 7.48%. Materials conferred with elite subunits of 1, 2~*, 13+16, 14+15, 17+18 were selected and could be a source of desirable genes. Results from the diversity analysis showed that the diversity of wild emmer wheat were greater than of durum wheat in the Glu-A1 and Glu-B1 loci, as well as HMW-GS composition. One new subunit combinations type was found in wild emmer wheat materials GW-2 and GW-4.
