目的 评估所建立的可溶性细胞间黏附分子-1生物素-亲和素系统(BAS)时间分辨荧光免疫分析方法(sICAM-1 BSA-TRFIA)的技术指标及应用价值.方法 建立sICAM-1 BsA-TR-FIA,分析其检测曲线的性质参数,研究该方法的灵敏度、精密度和回收率、特异性,稳定性、平行性,以及与酶联免疫吸附试验分析法(ELlSA)间的相关性.结果 此法特异性较高,同批试剂连续6个月应用分析结果稳定,灵敏度为1.32 μg/L,批内和批间CV分别为4.24%和6.83%,平均回收率为99.87%,线性范围为(2.5~1 933)μg/L,与ELISA所测结果比较高度相关(r=0.939),与TNF-а、IL-8、IL-1、IL-2和IL-6等免疫分子无交叉反应.结论 sICAM一1 BSA-TRFIA是一种特异性强、稳定性好、灵敏度高、可测范围较广的检测方法,可以方便地应用于临床,为相关疾病的诊断和监测提供一项良好的实验诊断指标.

Objective To evaluate the methodology and clinical application about the newly es-tablished time-resolved fluoroimmunoassay(TRFIA), which has been used to detect soluble intercellu-lar adhesion molecule-1 (sICAM-1) captured by the surface BAS-biotin. Methods After sICAM-1BSA-TRFIA test system was established, the parameters of the detection curve were analyzed, and further to study its sensitivity, precision, recovery rate, specificity, stability, and parallelism. Then the correlation of the method and enzyme linked immunosorbent assay(ELISA) was investigated. Re-sults This method had higher specificity and stability than ELISA. After 6 months of successive ap-plication of the same batch of agents, the analytic results kept stable. The sensitivity was 1.32 μg/L.The intra- and inter-batch CV were 4. 24% and 6.83%, respectively. The average recovery rate was 99.87%. On the test curve, the linear ranger was 2. 5-1 933μg/L. The detection results were closely correlative with those with ELISA(r=0. 939). No cross reaction exist between the new method and FIA is characteristic of strong specificity, excellent stability, high sensitivity, and wide detection range. It can be widely applied and conveniently used in the clinic as a laboratory index on clinical diag-nosis and monitoring.
