目的: 探讨人脑星形细胞肿瘤组织中的细胞凋亡,以及细胞凋亡与其相关基因bcl-2、Bax表达之间的关系。方法:采用原位末端标记的TUNEL技术及免疫组织化学S-P法对30例星形细胞肿瘤手术标本进行检测,分别观察了肿瘤组织中凋亡细胞密度及bcl-2、Bax基因的表达。结果:星形细胞肿瘤组织中普遍存在细胞凋亡现象,凋亡细胞密度随肿瘤恶性度的升高,呈逐渐降低的趋势(t=2.77,P<0.01);bcl-2、Bax在星形细胞肿瘤组织中表达阳性率分别为50%和100%,其中bcl-2表达强度与凋亡细胞密度呈显著负相关(r=-0.922,P<0.001)。结论:细胞凋亡受抑在星形细胞肿瘤发生及间变过程中起重要作用;bcl-2基因表达对细胞凋亡的发生起抑制作用;Bax基因对细胞凋亡的调节可能存在更为复杂的机制。
目的: 探討人腦星形細胞腫瘤組織中的細胞凋亡,以及細胞凋亡與其相關基因bcl-2、Bax錶達之間的關繫。方法:採用原位末耑標記的TUNEL技術及免疫組織化學S-P法對30例星形細胞腫瘤手術標本進行檢測,分彆觀察瞭腫瘤組織中凋亡細胞密度及bcl-2、Bax基因的錶達。結果:星形細胞腫瘤組織中普遍存在細胞凋亡現象,凋亡細胞密度隨腫瘤噁性度的升高,呈逐漸降低的趨勢(t=2.77,P<0.01);bcl-2、Bax在星形細胞腫瘤組織中錶達暘性率分彆為50%和100%,其中bcl-2錶達彊度與凋亡細胞密度呈顯著負相關(r=-0.922,P<0.001)。結論:細胞凋亡受抑在星形細胞腫瘤髮生及間變過程中起重要作用;bcl-2基因錶達對細胞凋亡的髮生起抑製作用;Bax基因對細胞凋亡的調節可能存在更為複雜的機製。
목적: 탐토인뇌성형세포종류조직중적세포조망,이급세포조망여기상관기인bcl-2、Bax표체지간적관계。방법:채용원위말단표기적TUNEL기술급면역조직화학S-P법대30례성형세포종류수술표본진행검측,분별관찰료종류조직중조망세포밀도급bcl-2、Bax기인적표체。결과:성형세포종류조직중보편존재세포조망현상,조망세포밀도수종류악성도적승고,정축점강저적추세(t=2.77,P<0.01);bcl-2、Bax재성형세포종류조직중표체양성솔분별위50%화100%,기중bcl-2표체강도여조망세포밀도정현저부상관(r=-0.922,P<0.001)。결론:세포조망수억재성형세포종류발생급간변과정중기중요작용;bcl-2기인표체대세포조망적발생기억제작용;Bax기인대세포조망적조절가능존재경위복잡적궤제。
Aim: To investigate the status of apoptosis and the relationship between apoptosis and the expression of its related-genes bcl-2 and Bax in tissues with astrocytic tumor. Methods: TUNEL and immunohistochemical S-P technique were used to determine apoptotic cell density (AD), bcl-2 and Bax expression in thirty cases of cerebral astrocytic tumor. Results: Apoptosis was very common in all the thirty cases.AD in astrocytoma tissues presented a significant decreasing trends with the tumor grade of maligance increasing. Meanwhile, it was found that bcl-2 and Bax positive expression in astrocytic tumor were 50% and 100% respectively. The extent of bcl-2 expression had a significant negative corrlation with AD (r=-0.922, P<0.001). No relationship was found between Bax expression and AD (P>0.05). Conclusion: The inhibition of cell apoptosis may play an important role in astrocytic tumorigenesis and its anaplastia. The product of bcl-2 gene expression may inhibit cell apoptosis in tissues of astrocytic tumor. Bax gene may regulate tumor cell apoptosis via more complicated mechnism, which remains to be elucidated by further research.
