目的探讨复制蛋白A1(RPA1)在人食管癌组织中的表达及其临床意义.方法采用逆转录-聚合酶链反应(RT-PCR)和免疫组织化学染色法检测40例食管癌组织和癌旁组织中RPA1的mRNA及蛋白.结果 RPA1的mRNA及蛋白在食管癌组织和癌旁组织中均有表达,食管癌组织中RPA1表达水平明显高于相应的癌旁组织(P＜0.05),食管癌组织中RPA1表达水平与患者年龄、性别以及肿瘤大小无明显相关,而与肿瘤的浸润深度以及淋巴结转移明显相关.结论食管癌组织中RPA1的表达与其进展及转移密切相关.

Objective To study the expression and significance of replication protein A1 ( RPA1 )in esophageal carcinoma.Methods The expressions of RPA1 mRNA and protein was detected by using reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry in 40 esophageal squamous cell carcinoma samples and the tumor-adjacent normal tissues from the same cases.Results RPA1 mRNA and protein were expressed in esophagus carcinoma and the tumor-adjacent normal tissues.The RPA1 mRNA and protein expression levels in esophagus carcinoma tissues were significantly higher than those in the tumor-adjacent normal tissues (P ＜ 0.05).RPA1 mRNA and protein levels in patients with esophagus carcinoma were not correlated with with age,sex,and the site of the tumor,but with invasive depth and lymph node metastasis.Conclusion RPA1 expression in esophagus carcinoma tussues may be closely associated with the progression and metastasis and RPA1 may be used as a valuable target for early diagnosis.
