利用乳酸杆菌分离培养基(SL)对犊牛瘤胃内的乳酸杆菌进行分离,对分离出的乳酸杆菌进行革兰氏染色、生化反应及16S rRNA同源性分析鉴定.对分离鉴定的干酪乳杆菌提取其基因组,根据Genbank 发表的干酪乳杆菌耐酸相关基因(ffh基因)设计1对引物进行PCR,利用pMD18-T载体进行克隆,构建重组质粒,进行PCR及测序鉴定,获得耐酸基因ffh,为下一步研制瘤胃微生态制剂及构建瘤胃内耐酸的乳酸分解基因工程菌奠定了基础.
利用乳痠桿菌分離培養基(SL)對犢牛瘤胃內的乳痠桿菌進行分離,對分離齣的乳痠桿菌進行革蘭氏染色、生化反應及16S rRNA同源性分析鑒定.對分離鑒定的榦酪乳桿菌提取其基因組,根據Genbank 髮錶的榦酪乳桿菌耐痠相關基因(ffh基因)設計1對引物進行PCR,利用pMD18-T載體進行剋隆,構建重組質粒,進行PCR及測序鑒定,穫得耐痠基因ffh,為下一步研製瘤胃微生態製劑及構建瘤胃內耐痠的乳痠分解基因工程菌奠定瞭基礎.

The SL medium was used to isolate Lactobacillus casei. The Gram staining, biochemistry I-dentification, morphology observation and 16S rRNA phylogenelic analysis were used to identify the bacteria. The genome of the identified Lactobacillus casei was extracted, and according to gene ffh from GenBank, a pair of primers were designed for PCR, then the gene was cloned into pMD 18-T vector to obtain the recombinant plasmid and detected by PCH and homology sequence analysis. The whole sequence of gene ffh was acquired. This work made preparation for the study of rumen microbial ecological agents and the acid-resistant genetic engineering degradation bacteria of lactic acid.
