研究利用高效液相色谱法快速分析新疆地产干果杏仁、苦杏仁和桃仁中苦杏仁苷的含量.采用Shim-pack VP-ODS(150mm×4.6mm,5μm)色谱柱,甲醇-水(20:80,V/V)为流动相,进样量为2μL,流速为1mL/min,柱温为40℃,在波长215nm处检测苦杏仁苷,并进行方法学考察.结果表明,利用此方法能有效分离并快速分析干果中苦杏仁苷含量,建立的回归方程为Y=4.86244×10~(-6)X-0.0129273,具有良好的线性和重复性,可作为在线检测干果中苦杏仁苷的定量手段.

A rapid high performance liquid chromatographic (HPLC) method was presented for determining amygdalin in Semen Armeniacae Amarum,bitter almond and Semen Persicae.The chromatographic separation was performed on a Shim-pack VPODS column (150 mm×4.6 mm,5μm) with a mobile phase composed of methanol and water (20∶80,V/V) at a flow rate of 1 mL/min,the column temperature was set at 40℃,and the analyte was detected at 215 nm wavelength.Results showed that mygdalin was separated well and analyzed effectively by the method.There was a good linearity between relative peak area (X) and amygcalin concentration (Y),and the linear regression equation was Y=4.86244×10~(-6)X-0.0129273.Methodological investigations demonstrated the advantages of this method,including good repeatability and high precision,which benefit its application to rapid online determination of amygdalin in dried nuts.
