目的 探讨大黄酸对脑出血(ICH)大鼠神经损伤和细胞凋亡的影响.方法 在成年SpragueDawley大鼠尾状核注入collagenase IV(0.05 U/0.5 uL)诱导ICH,假手术大鼠在相同位置注入相同剂量的氯化钠注射液.大鼠术后分别在3 h,6 h、12 h腹腔注射大黄酸注射液(70 ms/kg)或相同剂量的氯化钠注射液.Western blot法测定半胱氨酸天冬氨酸蛋白酶-3(caspase-3)活性,TUNEL标记凋亡细胞,阿朴吗啡诱导的旋转行为评价神经功能缺损.结果 大鼠脑出血后3 h和6 h给予大黄酸注射液明显抑制casepase-3的激活(P<0.001),减少TUNEL阳性细胞数(P<0.05),减少阿朴吗啡诱导的旋转次数(P<0.05);脑出血后12 h给药无效.结论 脑出血后早期给予大黄酸,可能通过阻断凋亡通路起到神经保护作用.
目的 探討大黃痠對腦齣血(ICH)大鼠神經損傷和細胞凋亡的影響.方法 在成年SpragueDawley大鼠尾狀覈註入collagenase IV(0.05 U/0.5 uL)誘導ICH,假手術大鼠在相同位置註入相同劑量的氯化鈉註射液.大鼠術後分彆在3 h,6 h、12 h腹腔註射大黃痠註射液(70 ms/kg)或相同劑量的氯化鈉註射液.Western blot法測定半胱氨痠天鼕氨痠蛋白酶-3(caspase-3)活性,TUNEL標記凋亡細胞,阿樸嗎啡誘導的鏇轉行為評價神經功能缺損.結果 大鼠腦齣血後3 h和6 h給予大黃痠註射液明顯抑製casepase-3的激活(P<0.001),減少TUNEL暘性細胞數(P<0.05),減少阿樸嗎啡誘導的鏇轉次數(P<0.05);腦齣血後12 h給藥無效.結論 腦齣血後早期給予大黃痠,可能通過阻斷凋亡通路起到神經保護作用.
목적 탐토대황산대뇌출혈(ICH)대서신경손상화세포조망적영향.방법 재성년SpragueDawley대서미상핵주입collagenase IV(0.05 U/0.5 uL)유도ICH,가수술대서재상동위치주입상동제량적록화납주사액.대서술후분별재3 h,6 h、12 h복강주사대황산주사액(70 ms/kg)혹상동제량적록화납주사액.Western blot법측정반광안산천동안산단백매-3(caspase-3)활성,TUNEL표기조망세포,아박마배유도적선전행위평개신경공능결손.결과 대서뇌출혈후3 h화6 h급여대황산주사액명현억제casepase-3적격활(P<0.001),감소TUNEL양성세포수(P<0.05),감소아박마배유도적선전차수(P<0.05);뇌출혈후12 h급약무효.결론 뇌출혈후조기급여대황산,가능통과조단조망통로기도신경보호작용.
Objective The purpose of this investigation was to investigate the neuroprotective effects of rhubarb for ICH,as well as its mechanism.Methods ICH was produced in adult Spargue-Dawley rats by injection of collagenase IV(0.05U/0.5uL).Intraperitoneal injection of rhubarb (70mg/kg) or saline,was started at 3,6 or 12 hours post-ICH respectivdy.Casepase-3 activity.TUNEL and neurological behavior function were performed 24hours after ICH.Results Rhubarb siven at 3 or 6 hours can inhibit casepase-3 activity(P<0.001),reduce TUNEL positive cells(P<0.05) and attenuate apomorphine-induced rotation(P<0.05) at 24 hours after ICH.However,the animals which were treated 12 hours showed no improvement.Conclusion Rhubarb may be a potential drug for ICH patients for its possible effect of inhibiting apoptosis.
