采用KPR和R2培养基进行水稻原生质体培养。日本晴、楚粳8号、云粳9号、大白谷和元江普通野生稻先后获得了原生质体培养的成功,除日本晴外,其余品种均为首次报道。品种间绿苗再生率有显著差异,日本晴最高,大白谷最低。同一品种不同的愈伤组织间,分化率有很大差异,有的愈伤组织极易分化出绿苗,有的不能再生出绿苗,说明分化前的预培养非常重要。N6分化培养基的再生绿苗率极显著地高于MS。配方为NMaizeResearch Institute,Sichuan Agricultural University,Ya'an 6 25014,China+葡萄糖20g/L+蔗糖30g/L+IAA0.2mg/L+6-BA0.5mg/L+Agarose-110g/L的培养基,分化率最高,按初次接种的愈伤组织块计,每块平均再生绿苗1.4苗。酶解原生质体量的多少,与愈伤组织在N6+1mg/L2,4-D的液体培养基中悬浮培养时间的长短有关,悬浮时间较长则易于获得较多的原生质体。日本晴的再生植株中,有两株从移栽到抽穗仅45d,有可能是极早熟的变异株,有些植株的粒型和稃毛也有明显变异,对鉴定和利用变异的植株具有现实意义。

Green plantlets were successfully gotten from protopl asts ofYunnan rice varieties,Chujing 8,Yunjing 9,Dabaigu and Yuanjiang common w ild rice and a Japanese variety,Nipponbare.Regeneration frequencies(RF) of green plantlets among varieties showed significant difference.Within one variety,some calli seemed to be very easily regenerated while other calli were difficult to g et green plantlets in spite of using many kinds of media.RF in medium N6 was muc h higher than that in medium MS.The yields of protoplasts were related to the ag es of suspension culture.Some plants derived from protoplast of Nipponbare showe d different seed shape,glume hairiness and mature time from the original plants. In these plants,genetic variation might have been induced during the period of in vitro culture.
