目的 研究NF-κB圈套寡核苷酸对SAH后脑血管痉挛(CVS)的防治作用.方法 利用兔枕大池二次注血模型,在二次注血前枕大池分别注入NF-κB诱骗寡核苷酸、乱序寡核苷酸与脂质体的混合物,利用DSA测定兔基底动脉直径,了解CVS的情况,并以常规HE染色、免疫组化和原位杂交等方法观察兔基底动脉管壁形态以及NF-κB在管壁上的表达.结果 统计学结果显示,NF-κB诱骗寡核苷酸组血管狭窄的程度、形态学改变以及NF-κB在管壁上的表达较乱序寡核苷酸组和对照组明显减轻.结论 NF-κB圈套作为一种新的治疗模式,对CVS的防治有良好的效果.

목적 연구NF-κB권투과핵감산대SAH후뇌혈관경련(CVS)적방치작용.방법 이용토침대지이차주혈모형,재이차주혈전침대지분별주입NF-κB유편과핵감산、란서과핵감산여지질체적혼합물,이용DSA측정토기저동맥직경,료해CVS적정황,병이상규HE염색、면역조화화원위잡교등방법관찰토기저동맥관벽형태이급NF-κB재관벽상적표체.결과 통계학결과현시,NF-κB유편과핵감산조혈관협착적정도、형태학개변이급NF-κB재관벽상적표체교란서과핵감산조화대조조명현감경.결론 NF-κB권투작위일충신적치료모식,대CVS적방치유량호적효과.
Objective To evaluate the effect of NF-kappaB decoy oligodeoxynucleotide as means of prevention and treatment on CVS after SAH.Method The models of subarachnoid hemorrhage(SAH)and CVS were successfully made via double blood injection into cisterna magna.A double-stranded oligo-DNA containing the rabbit NF-κB and a scrambled oligo-DNA were transferred into the subarachnoid space of the rabbits through the cerebrospinal fluid via a cationic liposome delivery system before the second iniection.DSA was performed to detect the diameter of BA which weas used to evaluate the degree of CVS.Morphological methods, immunohistochemistry and hybridization in situ were employed to observe the morphological changes of basilar artery and the dynamic expression of NF-κB in basilar artery walls.Results The stenosis degree of the BA,morphological changes and the expression of NF-κB were more slight in decoy groups than those in scrambled oligo-DNA group and control group.Conclusions NF-κB decoy strategy,as a novel anti-inflammatory means,showed good effect for prevention and treatment on CVS after SAH.
