Micromachined capillary electrophoresis systems with integrated sample injector have been fabricated on glass chips using standard photolithography and chemical wet etching techniques. Experiments have been done to find proper ways of obtaining regular and smooth channels, and the performance of the chip was evaluated. Voltages of 4 kV could be applied over an 20 mm long capillary channel with sufficient dissipation of the Joule heat generated (1.12 W/cm). Electroosmotic flow was used to "load" the sample into the microchip and to "pump" the mobile phase during the experiments. Using laser-induced fluorescence detection and CCD imaging, the mixture of FITC-OH and FITC-labeled amino acid were injected and separated. Two methods of sample introduction ("floating" and "pinched") were evaluated. The results showed that applications of potentials to the intersecting channels could cut the sample plug clearly during the process of separation which made the tailing unserious. Frequent flushing of channels with alkaline solution and water was demonstrated to be important to keep the reproducibility of sample loading and separation.
