目的 观察香烟烟雾暴露大鼠气道炎症模型血清和支气管肺泡灌洗液(BALF)中白介素17(IL-17)、IL-6的变化,以探讨其可能在气道炎症中所起的作用.方法 40只健康清洁级雄性Wistar大鼠随机分为四组:12周正常对照组(C1组)、24周正常对照组(C2组)、12周香烟烟雾暴露组(S1组)、24周香烟烟雾暴露组(S2组),每组10只.烟熏法复制大鼠气道炎症的动物模型.收集BALF行细胞计数和分类计数;用酶联免疫吸附试验法检测各组大鼠模型血清和BALF上清液中IL-17、IL-6的浓度.结果 ①与C1、C2组相比,S1、S2组BALF中细胞总数,各类细胞数都有不同程度增高,S2组增高尤其明显.②与C1、C2组比较,S1、S2组血清和BALF中IL-17浓度升高(P<0.05),与S1组相比,S2组IL-17浓度明显升高(P<0.01);与C1组比较,S1组血清IL-6浓度未见明显升高(P>0.05),但BALF中升高明显(P<0.05),与C2组比较,S2组血清和BALF中IL-6浓度均升高(P<0.05),与S1组比较,S2组血清与BALF中IL-6浓度也明显升高(P<0.05).③香烟烟雾暴露组外周血IL-17与IL-6水平呈正相关(r=0.463,P=0.040),而BALF中IL-17与IL-6水平无明显相关(r=0.269,P=0.252),且与BALF细胞涂片中的淋巴细胞、中性粒细胞和单核巨噬细胞均无相关性.结论 香烟烟雾暴露致使大鼠气道炎症模型血清和BALF中IL-17、IL-6水平增高,两者可能共同参与了COPD的发生与发展过程.

Objective The aim to investigate the change of interleukin-17(IL-17)and IL-6 in surem and bronchoalveolar lavage fluid(BALF)in cigarette exposure rats,to observe the effection of this two factors on an airway inflammation.Methods 40 male Wistar rats were randomly divided into four groups:12 weeks control group(C1 group),24 weeks control group(C2 group),12 weeks smoke-exposure group (S1 group),24 weeks smoke-exposure group(S2 group),10 rats each group.At the 12 week and 24 week,the groups were sacrificed.Total and differential ceils were counted in BALF for observed,enzyme linked immunosorbent assay(ELISA)was used to detect the 1evels of IL-17 and IL-6 in surem and BALF.Results ①Compared with the C1 and C2 groups,the levels of total and differential cell counts were increased in the cigarette exposure groups,especially in the S2 group.②Compared with C1 and C2 groups,the level of IL-17 not only in surem but in BALF were ignificantly increased in S1 and S2 groups (P<0.05).It was significantly increased in S2 group compared with the S1 group too (P<0.01).Compared with the C1 group,IL-6 in surem in the S1 group was not changed(P>0.05),but it's significantly increased in BALF.Compared with the C2 group,level of IL-6 increased both in surem and BALF,it's also ignificantly increased in S2 group compared with S1 group.③There was a positive correlation between IL-17 and IL-6 in serum but not in BALF in cigarette exposure groups.otherwise,the IL-17 from BALF in cigarette exposure groups was failed to correlate with neutroeyte,lymphocyte,monocyte.Conclusions The levels of IL-17 and IL-6 were increased in serum and BALF in airway inflammation model with cigarette exposure rats,they maybe related with the occurrence and development of COPD.
